4.4.1 Starting a run using default settings<br />

After the <strong>CombiFlash</strong> Torrent module has been automatically<br />

primed (See AUTO PRIME/VALVE WASH [4.3.7]) and a RediSep Rf<br />

column has been inserted, the module is ready to perform a<br />

run using the default settings. These default settings are<br />

preset to the most common configurations used by chemists,<br />

and have been optimized for the use of RediSep Rf columns.<br />

The default settings will:<br />

• collect all fluid in the fraction collector rack<br />

• differentiate between UV (254 nm) vs. non-UV absorbing<br />

fluid and place the fluids into separate tubes<br />

• cut UV absorbing peaks based on slope detection or level<br />

threshold.<br />

To start the run from the PeakTrak Main window:<br />

1. Enter a sample name. If you choose not to, PeakTrak will<br />

enter the date and time as the sample name when you start<br />

the run.<br />

2.Review the PeakTrak Main window settings. If you want to<br />

change any settings, refer to the Editing a Method on-line<br />

help topic.<br />

3.Click the Play button.<br />

4.Select the sample Loading Type from the list. The Loading<br />

Type should support the sample injection technique you<br />

intend to use (see SAMPLE INJECTION METHODS [ON-LINE]).<br />

Possible sample loading types are:<br />

• Solid (Pause) – Select this option if you intend place<br />

the sample into a solid sample load cartridge, but<br />

have not yet prepared the sample. When you click the<br />

run button, the module will perform a column<br />
