Section 4 <strong>User</strong> Help Reference<br />
Note: If both Slope and Threshold peak detection<br />
methods are checked, PeakTrak considers a peak to<br />
be present when any one condition is met. This logical<br />
OR operator means that PeakTrak will cut a peak<br />
when either the Slope condition is true, or when the<br />
Threshold condition is true.<br />
• All Wavelength Detection — Enable this option to<br />
detect peaks within a user-selected range of<br />
wavelengths. When enabled, click the Details button to<br />
configure additional settings. These settings include the<br />
slope-based and peak width options described in 1 and<br />
2 above. Additionally, you can type or select the<br />
minimum and maximum wavelengths limits in<br />
nanometers.<br />
• External Detector — Enable this option to use a 0 to 1<br />
volt analog signal from an external detector. When<br />
enabled, click the Details button to configure additional<br />
settings. These settings include the slope-based and<br />
peak width options described in 1 and 2 above.<br />
PeakTrak will use its internal algorithms to detect and<br />
cut peaks based on the analog input signal. Refer to the<br />
External Detector instruction sheet for electrical and<br />
plumbing requirements.<br />
• Show Ratio — If using two of the above peak detection<br />
options, check this option to display a ratio of the<br />
selected wavelengths. The ratio trace is often a useful<br />
indicator of purity. Refer to the MONITORING THE<br />
WAVELENGTH RATIO [4.4.7] topic for more details on this<br />
feature.<br />
4.3.3 Gradient Optimizer<br />
Use this window to automatically generate a gradient curve<br />
from the results of two TLC plates. Follow the on-screen<br />
instructions, then enter valid TLC data in the Gradient<br />
Optimizer window. Click the OK button and the Method<br />
Editor window will display the new gradient curve.<br />
When using the Gradient Optimizer, PeakTrak may insert an<br />
isocratic hold into the generated gradient. Furthermore,<br />
PeakTrak may automatically extend the isocratic hold during<br />
the run to increase peak resolution.<br />
