<strong>Column</strong> Information and Sample Run<br />

Proper conditioning and use of Teledyne <strong>Isco</strong>’s<br />

<strong>RediSep</strong> ® <strong>Alumina</strong> <strong>Neutral</strong> functionalized columns will<br />

help to improve column consistency and reliability. This<br />

instruction sheet provides a sample run that may be<br />

used as a column performance benchmark.<br />

<strong>Column</strong> <strong>Care</strong><br />

Initial <strong>Column</strong> Conditioning<br />

The column should be equilibrated with a minimum<br />

of three column volumes. Normally columns are equilibrated<br />

with the initial conditions of the gradient. The<br />

equilibration can be written into the method if desired.<br />

Equilibration Between Runs<br />

<strong>RediSep</strong> ® <strong>Alumina</strong> <strong>Neutral</strong> columns are intended for<br />

single-use only. However, repeat use of the column may<br />

be possible assuming that all sample was eluted off the<br />

column during the previous run. Three to five column<br />

volumes are a sufficient amount of equilibration<br />

between runs. If baseline drift is noticeable early in the<br />

run then more equilibration may be needed between<br />

runs.<br />

Sample Run<br />

The sample run purifies a 1:1 mixture of 6-Chlorochromone<br />

and Propyl 4-hydroxybenzoate (Figure 1)<br />

using the parameters in Table 1. The chromatogram is<br />

illustrated in Figure 2.<br />
