Column Information and Sample Run
Proper conditioning and use of Teledyne Isco’s
RediSep ® Alumina Neutral functionalized columns will
help to improve column consistency and reliability. This
instruction sheet provides a sample run that may be
used as a column performance benchmark.
Column Care
Initial Column Conditioning
The column should be equilibrated with a minimum
of three column volumes. Normally columns are equilibrated
with the initial conditions of the gradient. The
equilibration can be written into the method if desired.
Equilibration Between Runs
RediSep ® Alumina Neutral columns are intended for
single-use only. However, repeat use of the column may
be possible assuming that all sample was eluted off the
column during the previous run. Three to five column
volumes are a sufficient amount of equilibration
between runs. If baseline drift is noticeable early in the
run then more equilibration may be needed between
runs.
Sample Run
The sample run purifies a 1:1 mixture of 6-Chlorochromone
and Propyl 4-hydroxybenzoate (Figure 1)
using the parameters in Table 1. The chromatogram is
illustrated in Figure 2.
