ranging from 2,150 to 2,800 meters. Rasaut (also called rasaunt or rasanjana) is a<br />

useful concentrated extract obtained from roots of this plant and is used in curing<br />

many human ailments. Moreover, the plant has hepatoprotective, antiturnour,<br />

sedative and wound healing properties. The chief constituent of the roots and stem<br />

bark of B. aristata is an alkaloid Berberine, which is reported to be I responsible<br />

for most of the biological activity of the plant. A study was conducted to identify<br />

high berberine yielding plants/populations and to assess the seed germination<br />

potential of the plant growing in Himachal Pradesh. Berberine content was<br />

estimated using High Performance Liquid Chromatography (HPLC). The chemical<br />

analysis showed maximum berberine content of 2. 81 per cent in sample No. 30<br />

collected from Shimla district. Effect of different treatments on seed germination<br />

of the plant was also studied under laboratory conditions. The seed germination<br />

studies revealed that maximum germination (93. 50%) was recorded in seeds<br />

treated with hot water fot 24 hours. In this paper variation in berberine content and<br />

seed germination potential of B. aristata growing in Himachal Pradesh have been<br />

discussed.<br />

Disintegration of microbial cells is a necessary first step for the purification or<br />

extraction of intracellular enzymes and organelles. With increasing use of intracellular<br />

microbial materials in industry and medicine, the cell disruption unit operation<br />

is gaining in importance. L-asparaginase (E. C. 3. 5. 1. 1.) is a therapeutic enzyme<br />

and extensively used in the treatment of acute lymphoblastic leukemia. L-asparaginase<br />

is widely distributed among microorganisms, plants and animals. Due to the<br />

difficult extraction procedure to obtain the enzyme from plants and animals,
