CHAPTER 2: Material <strong>and</strong> Methods<br />

8 ml <strong>of</strong> PBS - (Gibco). The rema<strong>in</strong><strong>in</strong>g volume <strong>of</strong> 0.05% work<strong>in</strong>g solution was stored at 4°C <strong>and</strong><br />

used or discarded with<strong>in</strong> one week.<br />

2.1.1.4- Miscellaneous Culture Components, Solutions <strong>and</strong> Materials<br />

Bov<strong>in</strong>e Serum Album<strong>in</strong> (BSA) (Sigma-Aldrich) [Cat No: A8806] was used <strong>in</strong> reconstitution <strong>of</strong> FGF-<br />

2. To prepare a solution conta<strong>in</strong><strong>in</strong>g BSA, powder was weighed, allowed to dissolve undisturbed <strong>in</strong><br />

the solvent for 10-15 m<strong>in</strong>utes, <strong>and</strong> filtered through a 0.22 µm syr<strong>in</strong>ge filter unit.<br />

Capillaries, glass (Harvard Apparatus, Holliston MA, USA) [Catalog number 30-0039] were pulled<br />

over a low Bunsen burner flame to generate f<strong>in</strong>ely edged tips for cutt<strong>in</strong>g hESC colonies.<br />

Dimethylsulfoxide (DMSO) (Sigma-Aldrich, Ste<strong>in</strong>heim, Germany) [Cat No: D2650] was used at a<br />

f<strong>in</strong>al concentration <strong>of</strong> 10% (v/v) <strong>in</strong> freez<strong>in</strong>g medium for cryostorage <strong>of</strong> cell l<strong>in</strong>es <strong>in</strong> liquid nitrogen.<br />

Distilled water (Gibco) [Cat No: 15230-162]<br />

Fetal calf serum (FCS) used for MEF <strong>and</strong> hESC culture was batch tested <strong>and</strong> supportive batches<br />

were ordered <strong>in</strong> bulk to provide reagent for up to several years. Different types <strong>of</strong> FCS were used<br />

for specific tissue applications: for MEF culture JRH Biosciences FCS [Cat No: 12003-500N] <strong>and</strong><br />

CSL Biosciences FCS (Parkville VIC, Australia) [Cat No: 09702301] while for hESC culture<br />

Hyclone FCS (Logan UT, USA) [Cat No: SH30070-03] <strong>and</strong> JRH Biosciences FCS (Brooklyn VIC,<br />

Australia), [Cat No: 12203-100M].<br />

Fugene 6 Transfection Reagent (Roche Applied Science) [Cat No: 11 814 443 001] was obta<strong>in</strong>ed<br />

<strong>in</strong> 1 ml volume <strong>and</strong> stored at 4°C.<br />

Genetic<strong>in</strong> 50 mg/ml (Gibco) [Cat No: 10131-035], also known as G418 sulfate, is an<br />

am<strong>in</strong>oglycoside used as a selective antibiotic for genetically modified cell l<strong>in</strong>es which conta<strong>in</strong> the<br />

neomyc<strong>in</strong> resistance gene. Work<strong>in</strong>g concentration <strong>in</strong> cell culture media was 500 µg/ml.<br />

Pasteur pipettes, glass disposable (Chase Scientific Glass, Rockwood, Tennessee) [No. 93, 9<br />

<strong>in</strong>ch length, fl<strong>in</strong>t glass] were packed <strong>in</strong>to metal canisters <strong>and</strong> baked at 180°C for 2 hours to<br />

sterilize. Glass Pasteur pipettes were attached to a vacuum tap with tub<strong>in</strong>g <strong>and</strong> used to aspirate<br />

media from tissue culture flasks.<br />

Mitomyc<strong>in</strong> C (Sigma-Aldrich, Ste<strong>in</strong>heim, Germany) [Cat No: M0503] was received as 2 mg<br />

lyophilized powder <strong>and</strong> stored at 4°C. A 0.5 mg/ml stock solution was made <strong>by</strong> add<strong>in</strong>g 4 ml <strong>of</strong><br />

sterile distilled water (Gibco) [Cat No: 15230-204] <strong>in</strong>to each Mitomyc<strong>in</strong> C vial <strong>and</strong> allowed to<br />

dissolve completely. The solution was filter sterilized us<strong>in</strong>g a pre-wet 0.22 µm filter <strong>in</strong>to a 15 ml<br />

Falcon tube. The tube was protected from light with alum<strong>in</strong>um foil <strong>and</strong> stored at 4°C. The work<strong>in</strong>g<br />

solution was used at 10 µg/ml <strong>in</strong> MEF culture media immediately before add<strong>in</strong>g to MEF.<br />
