Protecting Group Chemistry<br />
* Protection of alcohols Not used for protection of alcohols but used for the protection of phenols-<br />
Phase transfer (BOC2O, Bu4NHSO4 , NaOH, DCM) or BOC2O, DMAP. Cleavage with 3M HCl in dioxane.<br />
* Protection of amides The rate of reaction with BOC2O increases as the acidity of the amide NH<br />
increases. DMAP catalyst is used. JOC 1997, 62, 2070<br />
* Protection of amines Used extensively for amine protection. Not hydrolysed under basic conditions<br />
and is inert to many nucleophilic reagents. A common method for its introduction is (BOC) 2O, NaOH,<br />
water, r.t. Non aqueous conditions: (BOC) 2O, TEA, DCM are also used. Acidic cleavage e.g. HCl in<br />
MeOH, TFA, TsOH. Imidazoles are also protected under phase transfer conditions or via NaH<br />

* Protection of thiols Protects thiols, both amine and thiol of cysteine is simultaneously protected.<br />
Treatment with HCl in EtOAc selectively removes the S-Boc group. JOC 1994, 59, 3216<br />
