ALELNA VARIJABILNOST U LOKUSU RHT8 GENA U<br />

NOVOSADSKOJ KOLEKCIJI PŠENICE I NJEN ZNAÈAJ U<br />

OPLEMENJIVAÈKIM PROGRAMIMA<br />

Visina stabljike pšenice je sloena osobina uslovljena velikim brojem gena kao i<br />

uticajima spoljašnje sredine. Korišæenje gena koji redukuju visinu stabljike, a poveæavaju<br />

prinos je jedna od glavnih strategija u dobijanju visoko prinosnih sorti hlebne pšenice.<br />

Do sada je identofikovano 21 major gena koji redukuju visinu pšenice, a od kojih se Rht8<br />

gen najèešæe koristi u oplemenjivaèkim programima u junoj Evropi.<br />

Sa ciljem da se utvrdi odgovor 350 genotipova pšenice iz novosadske kolekcije na<br />

egzogeno dodatu giberelinsku kiselinu, primenjen je GA test. Priblizno 22% od 350<br />

genotipova nije reagovalo izduivanjem ponika na egzogeno dodatu giberelinsku<br />

kiselinu, a 78% jeste. Alelna varijabilnost na Rht8 lokusu ovih genotipova utvðena je<br />

analizom mikrosatelitnog lokusa MR 21, koji je blisko vezan sa Rht8 lokusom. Nakon<br />

toga, ispitivan je efekat razlièitih alela na agronomske karakteristike i utvrðena je<br />

znaèajna veza izmeðu alelne varijabilnosti na Rht8 lokusu sa jedne strane i visine<br />

stabljike i komponenti prinosa sa druge strane.<br />

U radu su dati rezultati GA testa i molekularne analize, sa specijalnim naglaskom na<br />

potencijalnoj upotrebi ovih podataka u oplemenjivaèke svrhe.<br />

ALLELIC VARIATION AT THE DWARFING GENE RHT8 LOCUS<br />

IN NOVI SAD WHEAT COLLECTION AND ITS SIGNIFICANCE IN<br />

BREEDING PROGRAMMES<br />

The control of plant height in wheat is known to be complex, being determined by many<br />

genes, and subject to environmental effects. The use of dwarfing genes to reduce plant<br />

height and improve yield potential has been one of the major strategies in breeding modern,<br />

high yielding bread wheat cultivars. So far, 21 major genes for reducing plant height<br />

have been determined, among which Rht8 is most frequently used in wheat breeding programs<br />

in Southern Europe.<br />

In order to determine the respond of 350 genotypes from Novi Sad Wheat Core Collection<br />

to exogenously applied Gibberellic Acid, GA test was performed. Approximately,<br />

22% of 350 genotypes have shown to be GA-insensitive and 78% to be GA-sensitive. A<br />

wheat microsatellite locus MR 21, which is closely linked to Rht8 locus, was used to<br />

screen these wheat genotypes to assess the variation at Rht8 locus. Afterwards, effects of<br />

different alleles on agronomic traits were analyzed and significant relation was detected<br />

between allelic variation at Rht8 locus at one side and plant height and grain yield and its<br />

components on the other side.<br />

For both GA test and molecular screening, obtained results are discussed in this paper,<br />

with special emphasis on potential use of the data for breeding purposes.
