GENETIÈKA KARAKTERIZACIJA LOKALNIH POPULACIJA<br />

KUKURUZA POMOÆU SSR MARKERA<br />

Banka gena Instituta za kukuruz «Zemun Polje» poseduje preko dve hiljade lokalnih<br />

populacija kukuruza, klasifikovanih i okarakterisanih na morfološkom nivou. Da bi se<br />

omoguæila preciznija karakterizacija i izbegla pojava potencijalnih duplikata u kolekciji<br />

neophodna je analiza postojeæih genotipova primenom molekularnih markera.<br />

U ovom radu analizirana je 21 lokalna populacija kukuruza pomoæu 30 SSR markera.<br />

Korišæena je metoda grupnih uzoraka, gde je svaka populacija predstavljena sa po 30<br />

biljaka. Elektroforetsko razdvajanje amplifikovanih fragmenata je raðeno na agaroznom i<br />

denaturišuæem poliakrilamidnom gelu. Poliakrilamidni gelovi su bojeni srebro-nitratom.<br />

Statistièka analiza dobijenih podataka je uraðena pomoæu NTSYSpc2 programskog<br />

paketa, a za izraèunavanje genetièkih distanci korišæena je «Modifikovana Roderova<br />

Distanca» na osnovu izraèunatih alelnih frekvencija unutar populacija.<br />

Rezultati i diskusija æe biti prikazani na posteru.<br />

GENETIC CHARACTERIZATION OF LOCAL MAIZE<br />

POPULATIONS WITH SSR MARKERS<br />

The genebank in Maize Research Institute «Zemun Polje» maintains the collectin of over<br />

2000 local maize populations classified and characterised at the morphological level.<br />

Molecular marker application would enable more precise characterisation and avoid occurrence<br />

of duplicate accessions, thus supporting the maintenance of the collection.<br />

Thirty SSR markers were used to analyse twenty one local maize populations. DNA<br />

pooling strategy was done with thirty plants representing each population. Amplified<br />

fragments were separated both on denaturing polyacrilamide and agarose gels. Silver<br />

staining was performed on polyacrilamide gels. NTSYSpc2 programe package was applied<br />

for statistical analysis of the results. Genetic distance was calculated using Modified<br />

Rogers Distance based on allelic frequency scoring of the bands within accessions.<br />

Results and discussion will be presented on the poster.
