AFLP ANALIZA VARIJABILNOSTI LOKALNIH POPULACIJA KUKURUZA<br />

Karakterizacija genetièkog diverziteta lokalnih populacija kukuruza u Banci gena<br />

Instituta za kukuruz «Zemun Polje» trebalo bi da optimizuje odravanje postojeæe<br />

kolekcije i olakša korišæenje datih genotipova. Primena molekularnih markera omoguæava<br />

procenu genetièkog diverziteta direktno na DNK nivou prevazilazeæi nedostatke<br />

morfoloških i biohemijskih markera.<br />

Devetnaest lokalnih populacija kukuruza je analizirano korišæenjem èetiri AFLP prajmer<br />

kombinacije. Primenjena je metoda grupnih uzoraka, pri èemu je svaka ispitivana<br />

populacija predstavljena sa po 30 biljaka. Nakon denaturišuæe poliakrilamidne elektroforeze<br />

gelovi su obojeni srebro-nitratom. Statistièka analiza podataka je uraðena pomoæu<br />

NTSYSpc2 programskog paketa, a genetièka distanca je izraèunata primenom akardovog<br />

koeficijenta na osnovu binarnog zapisa (1/0) prisustva/odsustva traka kod ispitivanih<br />

populacija.<br />

Rezultati i diskusija æe biti prikazani na posteru.<br />

AFLP ANALYSIS OF MAIZE LOCAL POPULATION VARIABILITY<br />

Genetic diversity characterisation of local maize populations from Maize Research Institute<br />

«Zemun Polje» genebank should optimize and facilitate their conservation and use.<br />

Molecular markers allow the estimation of genetic diversity directly at DNA level, thus<br />

overcoming the demerits of morphological and biochemical markers.<br />

Nineteen local maize populations were analysed with four AFLP primer combinations.<br />

Thirty individual plants were bulked to form DNA pooled samples for each population.<br />

Silver staining was applied after the denaturating polyacrilamide electrophoresis. Statistical<br />

analysis was performed using NTSYSpc2 programe package and genetic distances<br />

were estimated by Jaccard coefficient based on binary codes (1/0) of the bands for their<br />

presence/absence within each population.<br />
