ANTIGENOTOKSIÈNI EFEKAT TERPENA IZ ALFIJE<br />

Istraivanja su pokazala da aktivne supstance lekovitih i aromatiènih biljaka poseduju<br />

inhibitorni ili modulatorni efekat na genotoksiène agense. Cilj rada je detekcija<br />

antigenotoksiènog efekta terpena iz etarskog ulja alfije i izuèavanje mehanizama<br />

reparacije DNK koji su ukljuèeni, pomoæu prokariotskih i eukariotskih model sistema.<br />

Korišæena je baterija testova na E. coli K12 kojom se prati efekat na UV-indukovanu<br />

mutagenezu u reparaciono sposobnom, i eksciziono defektnom soju; spontane mutacije;<br />

SOS indukcija i intrahromozomalna rekombinacija. Kao dodatak, uraðen je mikronukleus<br />

test na humanim limfocitima ozraèenim zracima.<br />

Etarsko ulja alfije ima znatno manji efekat na smanjenje broja UV-indukovanih mutanata<br />

u uvrA - soju, nego u eksciziono sposobnom soju. Takav efekat je naðen i kod svih testiranih<br />

monoterpena iz njegovog sastava (tujon, 1,8-cineol i kamfor). U mutator mutS soju,<br />

nije uoèen znaèajan efekat na broj spontanih mutanata. tujon inhibira UV-indukovani<br />

SOS odgovor. Etarsko ulje alfije, 1,8-cineol i kamfor blago stimulišu intrahromozomalnu<br />

rekombinaciju u ozraèenom recA + soju, dok tujon nema znaèajnog efekta.<br />

Mikronukleus test je pokazao da etarsko ulje alfije ima slab radioprotektivni efekat.<br />

Rezultati dobijeni u ovom istraivanju, kao i podaci o antimutagenoj i antikancerogenoj<br />

aktivnosti monoterpena, ohrabruju dalja istraivanja etarskog ulja alfije kao<br />

potencijalnog agensa za prevenciju kancera.<br />

ANTIGENOTOXIC EFFECT OF TERPENOIDS FROM SAGE<br />

It has been shown that active substances from medicinal and aromatic plants possess inhibiting<br />

or modulating effect on environmental genotoxic agenta. This work is designed<br />

to detect antigenotoxic effects of terpenes from essential oil of sage, and to study the<br />

DNA repair mechanisms involved, using pro- and eukaryotic model systems.<br />

The battery of tests on E. coli K12 measuring UV-induced mutagenesis in excision repair<br />

proficient and deficient strains, spontaneous mutations, SOS induction and intrachromosomal<br />

recombination was used. In addition, micronucleus test was performed in -irradiated<br />

human limphocytes.<br />

Essential oil of sage was significantly less effective in inhibition of UV-induced mutations in<br />

uvrA - strain than in excision repair proficient strain. Similar effect is observed with all tested<br />

monoterpenes from essential oil ( thujone, 1,8-cineole and camphor). No significant effect<br />

on spontaneous mutagenesis was found in mutator mutS strain. thujone inhibit UV-induced<br />

SOS induction. Essential oil of sage, cineole and camphor mildly stimulate intrachromosomal recombination<br />

in UV-irradiated recA + strain, while thujone has no significant effect. Essential<br />

oil of sage exhibits mild radioprotective effect in micronucleus test.<br />

Our results, as well as the data on antimutagenic and anticarcinogenic activity of<br />

monoterpenes, encourage further investigation of essential oil of sage for potential use as<br />

cancer preventing agent.
