GENETIÈKA SLIÈNOST GENOTIPOVA SOJE DOBIJENA<br />

NA OSNOVU GENETIÈKIH MARKERA<br />

Genetièka osnova soje je veoma uska. Tradicionalno, genetièki diverzitet soje procenjivao<br />

se na osnovu morfoloških i agronomskih svojstava ili na informacijama o poreklu.<br />

Cilj ovog istraivanja je bio da se proceni informativnost izabranih genetièkih markera da<br />

bi se utvrdio genetièki diverzitet izmeðu 20 genotipova soje, kao i svrstavanje genotipova<br />

soje u odreðene grupe u zavisnosti od genetièke sliènosti.<br />

U ovom radu korisitli smo morfološke, proteinske i RAPD markereza utvrðivanje<br />

genetièke sliènosti.proteini semena su izolovani i razdvojeni PAA elektroforezom.<br />

Genomska DNK iz ovih genotipova okarakterisana je RAPD markerima korišæenjem 20<br />

izabranih prajmera. Na osnovu prisustva/odsustva traka na proteinskim<br />

gelovima/amplifikovanih DNK fragmenata izraèunati su koeficijenti sliènosti izmeðu<br />

parova genotipova. Za klaster analizu korišæen je UPGMA metod, a za neophodna<br />

proraèunavanja korišæen je NTSYS-pc program.<br />

Prema dobijenim rezultatima, genotipovi soje su svrstani u odgovarajuæe grupe. RAPD<br />

Markeri su detektovali viši nivo polimorfizma nego proteinski markeri. DNK markeri<br />

kao što su RAPD markeri su informativniji u proceni genetièkog diverziteta nego<br />

proteinski markeri.<br />

GENETIC SIMILARITY AMONG SOYBEAN GENOTYPES REVEALED BY<br />

GENETIC MARKERS<br />

The genetic diversity of cultivated soybean is very narrow. Traditionally, its estimation<br />

has been based on the differences in morphological and agronomic traits or pedigree information.<br />

The objective of this research were to evaluate informativeness of chosen genetic markers<br />

for estimating genetic diversity among 20 soybean genotypes, and assigning soybean<br />

genotypes into groups according to genetic similarity.<br />

In this study, we used morphological, protein and RAPD markers to validate their genetic<br />

relationship.Seed proteins were isolated and separated by PAA electrophoresis.Genomic<br />

DNA from these genotypes was characterized by RAPD markers with 20 selected<br />

decamer primers. On the basis of the presence/absence of bands in protein gels/amplified<br />

DNA fragments, coefficients of similarity were calculated between pairs of genotypes.<br />

The similarity matrix was submited for hierarchial cluster analysis of unweighted pair<br />

group using arithmetic average (UPGMA) method and necessary computation were performed<br />

using NTSYS-pc program.<br />

According to obtained results, soybean genotypes were assigned to correspoding groups.<br />

RAPD markers detected hogher level of polymorphism in soybean than protein markers.<br />

DNA markers such as RAPDs are more informative in estimating genetic diversity compared<br />

with protein markers.
