PRIMENA SSR MARKERA RADI UTVRÐIVANJA PRISUSTVA<br />

QTL-OVA ZA UKORENJAVANJE I SADRAJ ABA U LISTU KOD<br />

KOMPOZITNIH POPULACIJA KUKURUZA<br />

Tolerantnost kukuruza na sušu je kompleksna osobina, èija je ekspresija uslovljena<br />

aktivnošæu veæeg broja gena. Sadraj ABA u listu i ukorenjavanje su neke od<br />

fiziološko-morfoloških osobina koje utièu na ispoljavanje ove tolerantnosti, a èiji su<br />

QTL-ovi mapirani kod razlièitih genotipova kukuruza. Tako je, na primer, u segregirajuæoj<br />

populaciji ukrštanja Os420xIABO78, QTL sa major efektom za sadraj ABA u<br />

listu identifikovan na hromozomu 2 (bin 2.04), a za ukorenjavanje na hromozomu 1 (bin<br />

1.06).<br />

Cilj ovog rada je bio da se utvrdi da li se QTL-ovi sa major efektom detektovani u<br />

jednom genetipu mogu detektovati u nekom drugom genetièkom okruenju primenom<br />

molekularnih markera datog hromozomskog regiona. Za realizaciju navedenog cilja<br />

analizirano je 10 tolerantnih i 10 netolerantnih populacija kukuruza, korišæenjem 12 SSR<br />

prajmer kombinacije – dve na bin 1.05/ 1.06, a 10 na bin 2.04/2.05. Elektroforeza je<br />

raðena na denaturišuæem poliakrilamidnom gelu, a nakon razdvajanja amplifikovanih<br />

fragmenata DNK je bojena srebro-nitratom. Analizom elektroforegrama, na osnovu<br />

prisustva/odsustva traka, izraèunata je razlika u frekvenciji alela izmeðu tolerantnih i<br />

netolerantnih populacija. Rezultati i diskusija æe biti prikazani na posteru.<br />

SSR MARKER APPLICATION FOR DETERMINING PRESENCE<br />

OF QTLs FOR ROOTING AND LEAF ABA CONTENT IN COMPOSITE<br />

MAIZE POPULATIONS<br />

Maize drought tolerance is a complex trait with its expression determined by many<br />

genes. Leaf ABA content and rooting are some of physiological-morphological traits influencing<br />

drought tolerance, whose QTLs were mapped in different maize genotypes.<br />

For example, in a segregating population, derived from the cross Os420xIABO78, a QTL<br />

with a major effect for leaf ABA content was identified on chromosome 2 (bin 2.04) and<br />

for rooting on chromosome 1 (bin 1.06).<br />

The objective of this study was to determine if the QTLs with major effects detected in<br />

one genotype could also be detected in some different genetic background using molecular<br />

markers within a chromosome region harboring the QTLs in question. In order to fulfill<br />

the objective 10 drought tolerant and 10 drought non-tolerant maize populations were<br />

analyzed with 12 SSR primer combinations – two on bin 1.05/1.06 and 10 on bin<br />

2.04/2.05. Electrophoresis was run on a polyacrilamide denaturating gel and then silver<br />

stained. Allele frequency differences between tolerant and non-tolerant populations were<br />

calculated based on presence/absence of the bands on the gel. Results and discussion<br />

will be presented on the poster.
