GENETIÈKA KARAKTERIZACIJA RANIH HIBRIDA KUKURUZA<br />

PRIMENOM PROTEINSKIH I RAPD MARKERA<br />

Poznavanje genetièke varijabilnosti germplazme kukuruza je vano zbog planiranja<br />

programa selekcije, konzervacije germplazme per se. Proizvoðaèi kukuruza daju<br />

prednost uniformnosti useva koja se u veæini sluèajeva postie gajenjem inbred linija i<br />

dvolinijskih hibrida. Genetièka uniformnost ukljuèuje rizik od velikih gubitaka u<br />

prinosima zbog izuzetne osetljivosti ovakvih useva na stresne faktore sredine. Ovaj rizik<br />

se moe smanjiti gajenjem nesrodnih dvolinijskih hibrida. Zbog toga je postojanje<br />

informacije o genetièkoj varijabilnosti hibrida koji se gaje na poljima je takoðe vrlo<br />

vano. U ovom radu ispitivali smo informativnost proteinskih i RAPD markera za<br />

genetièku karakterizaciju hibrida i utvrðivanje njihove genetièke srodnosti. Kao materijal<br />

je izabrano 11 ranih hibrida kukuruza FAO 100-200 sa najvišim prinosom koji su<br />

pogodni za gajenje u brdsko-planinskim podruèjima. Proteini klice rastvorljivi u solima<br />

izolovani su i razdvojeni PAA elektroforezom. Amplifikacija genomske DNK ovih<br />

genotipova uraðena je sa 15 RAPD prajmera. Dobijeni rezultati kao prisustvo/odsustvo<br />

proteinske frakcije ili RAPD fragmenta prevedeni su u binarni kod (1,0). Za<br />

izraèunavanje koeficijenata sliènosti i UPGMA klaster metodu korišæen je NTSYS-pc<br />

programski paket. RAPD markeri su detektovali visok nivo polimorfizma meðu<br />

ispitivanim hibridima i informativniji su za procenu genetièke srodnosti od proteinskih<br />

markera.<br />

GENETIC CHARACTERIZATION OF EARLY MATURING MAIZE HYBRIDS<br />

OBTAINED BY PROTEIN AND RAPD MARKERS<br />

Knowledge on genetic diversity of maize germplasm is important for planning breeding<br />

programmes, germplasm conservation per se etc. The maize farmers prefer uniform crop<br />

fields, which in the majority of cases is attained by using inbred lines and single cross hybrids.<br />

This genetic uniformity may cause losts in yield bacause of genetic sucseptibility<br />

of material to environmental stress factors. This risk may be reduced by the use of unrelated<br />

single cross hybrids, so information on genetic variability of maize hybrids is very<br />

important. In our study we examined usefulness of protein and RAPD markers for characterization<br />

of maize hybrids and evaluation of their genetic relatedness. 11 early maturing<br />

ZP maize hybrids from groups FAO 100-200 were chosen for the analysis on the basis<br />

of their superior yield. The salt solubile proteins from embryo tissue were isolated and<br />

separated by gel electrophoresis (SDS-PAGE). The amplification of genomic DNA from<br />

embryo tissue was performed with 15 RAPD primers. Presense/absense of protein fractions<br />

and DNA fragments were transformed to binary data (1,0). The computing of binary<br />

data was performed using NTSYS-pc software including coefficients of similarities<br />

and UPGMA clustering. RAPD markers detected high level of polymorphism among hybrids<br />

and were more informative in assessing genetic relatedness between them.
