High-affinity receptor proteins with peptidyl-prolyl cis–trans<br />

isomerase activity that combine with such immunosuppressants<br />

as cyclosporin A, FK506, and rapamycin. They prevent<br />

the activity of rotamase by blocking conversion between<br />

cis and trans rotamers of the peptide and protein substrate<br />

peptidyl–prolylamide bond. Immunophilins are important<br />

in transducing signals from the cell surface to the nucleus.<br />

Immunosuppressants have been postulated to prevent signal<br />

transduction mediated by T lymphocyte receptors which<br />

blocks nuclear factor activation in activated T lymphocytes.<br />

Cyclophilin- and FK506-binding proteins represent immunophilins.<br />

Drug–immunophilin complexes are implicated in<br />

the mechanism of action of the immunosuppressant drugs,<br />

cyclosporin, FK506, and rapamycin.<br />

western blot is used to detect any possible proteins associated<br />

with the target protein. Used to isolate or detect antigens<br />

present in complex mixtures of proteins in solution.<br />
