Bacteroides immunity<br />

Bacteroides fragilis has a capsular polysaccharide that<br />

serves as a virulence factor that led to use of the polysaccharide<br />

as a vaccine in rats. The vaccine produced excellent<br />

antibody levels but did not prevent abscess formation.<br />

In the rat model, the findings strongly point to the role of<br />

T cells in the immune response to B. fragilis in abscess<br />

prevention. Murine studies confirmed the rat findings and<br />

demonstrated that immune T cells are antigen-specific.<br />

Abscess formation and prevention in response to B. fragilis<br />

are mediated by T cell mechanisms. Abscess formation<br />

requires a precursor-type T cell. Prevention of abscess<br />

requires T cells belonging to the suppressor phenotype<br />

that communicate via a small polypeptide factor. These<br />

cells are antigen-specific but not major histocompatibility<br />

complex (MHC)-restricted.<br />

BAFF<br />

B lymphocyte activating factor that is a member of the<br />

TNF family. A survival factor needed for the maturation of<br />

splenic transitional T1 B cells into transitional T2 B cells.<br />

BAFF is necessary for the appearance of mature B cells<br />

in the periphery. BAFF signaling is believed to activate<br />

NF-κB and induces Bcl-2 expression. Autoimmunity develops<br />

in mice when BAFF is overexpressed, and elevated<br />

serum levels of BAFF have been described in human<br />

autoimmune diseases. It is produced mainly by monocytes,<br />

macrophages, and dendritic cells, but not by B cells. Proinflammatory<br />

stimuli stimulate its expression by macrophages.<br />

It occurs as both a homotrimeric transmembrane<br />

molecule and in a soluble homotrimeric form. It binds to<br />

three receptors, BAFF-R, TACI, and BCMA.<br />
