2D-GE separates prote<strong>in</strong> mixture <strong>in</strong> accordance with two dist<strong>in</strong>ct properties. In<br />

the first dimension, prote<strong>in</strong>s are separated depend<strong>in</strong>g on their isoelectric po<strong>in</strong>t (pI, is<br />

known the pH at which a prote<strong>in</strong> has no net charge) disparity, whereas <strong>in</strong> the second<br />

dimension, the system uses the molecular weight difference. It means that prote<strong>in</strong>s are<br />

resolved accord<strong>in</strong>g to their molecular weight. Isoelectric focus<strong>in</strong>g (IEF) is coupl<strong>in</strong>g with<br />

sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) for prote<strong>in</strong><br />

separation, generally.<br />

Recently, the importance of 2D-GE system is rapidly <strong>in</strong>creas<strong>in</strong>g, as it not only<br />

can be comb<strong>in</strong>e with the most biological studies but also has a capacity to separate up to<br />

10000 prote<strong>in</strong> spots on one gel. Furthermore, it can monitorize more than 5000 prote<strong>in</strong>s<br />

simultaneously, hav<strong>in</strong>g nearly 2000 prote<strong>in</strong>s rout<strong>in</strong>ely, h<strong>in</strong>g<strong>in</strong>g upon the pore size <strong>in</strong><br />

acrylamide gels and pH gradient utilized. As compared to 1D-GE, it is more sensitive<br />

thanks to the properties of detection and quantification nearly 1 ng amount of prote<strong>in</strong><br />

per spot.<br />

By load<strong>in</strong>g a range of prote<strong>in</strong> markers whose molecular masses are already<br />

known <strong>in</strong> one of the l<strong>in</strong>e of the polyacrylamide gel, the molecular weights of the<br />

prote<strong>in</strong>s <strong>in</strong> the sample can be estimated.<br />
