The β-1, β-2 and β-5 subunits conta<strong>in</strong> the post-glutamyl peptidyl hydrolytic-like,<br />

tryptic-like, and chymotryptic-like proteolytic activities of the 26S proteasome,<br />

respectively (Groll et al., 1997) (Figure 1.9).<br />

Prote<strong>in</strong>s enter<strong>in</strong>g the 20S catalytic core are degraded <strong>in</strong> a progressive manner,<br />

result<strong>in</strong>g <strong>in</strong> the release of oligopeptides compos<strong>in</strong>g of three to twenty two am<strong>in</strong>o acids<br />

<strong>in</strong> length (Goldberg et al., 1997; Hershko and Ciechanover, 1998; DeMart<strong>in</strong>o and<br />

Slaughter, 1999; Delcros et al., 2003).<br />

1.2.7.3. Ubiquit<strong>in</strong>-Proteasome Pathway (UPP)<br />

The degradations process is chiefly mediated by the ubiquit<strong>in</strong>–proteasome<br />

pathway (UPP). The b<strong>in</strong>d<strong>in</strong>g of ubiquit<strong>in</strong> to the target prote<strong>in</strong>s from its ε-am<strong>in</strong>e of<br />

lys<strong>in</strong>e residues <strong>in</strong>volves a series of ATP-dependent enzymatic steps by E1 (ubiquit<strong>in</strong>-<br />

activat<strong>in</strong>g), E2 (ubiquit<strong>in</strong>-conjugat<strong>in</strong>g) and E3 (ubiquit<strong>in</strong>-ligat<strong>in</strong>g) enzymes. This<br />

comb<strong>in</strong>ation forms the poly-ubiquit<strong>in</strong>ated prote<strong>in</strong> which serves as substrate for the<br />

destruction of targeted prote<strong>in</strong> by the proteasome (Glickman and Ciechanover, 2002)<br />

(Figure 1.10).<br />
