Expression in Asbestos Lung Tissue<br />

Inhalation of asbestos increases the risk of lung cancer and pulmonary fibrosis. It is difficult to directly assess the<br />
distribution and content of inhaled particles in lung tissue sections. We showed that in-air micro particle induced X-ray<br />
emission (in-air micro-PIXE) system is useful for assessing the distribution and quantities of asbestos fibers and other metals<br />
in lung tissue comparing to immune-related cell localizations. Furthermore, the analysis using the system confirmed that<br />
asbestos induced apoptosis by upregulating Fas expression and also revealed the accumulation of CD163-expressing<br />
macrophages in the lungs of patients with asbestosis. By quantitative comparison of the area of Fas or CD163 expression<br />
and the Fas- or CD163-negative area in asbestos lung tissue, the harmful levels which caused the expression of Fas or CD163<br />
could be estimated on Si, Fe and Mg deposition. These results indicate that the system could be useful for investigating the<br />
pathogenesis of inhaled particle-induced immune reactions and for determining harmful levels of exogenous agents.<br />
アスベストは、天然の鉱物繊維で長さは約 8 m, 幅<br />
約 0.1 m、断熱性、耐火性、防音性、耐腐食性に優れ<br />
ており、建築用製材などに多く用いられてきた。しか<br />
し、アスベストの吸入は、肺線維症や肺ガンの原因で、<br />
発病までの潜伏期間が数 10 年と長いことから、「静か<br />
な時限爆弾」とも言われ、大変な社会問題となってい<br />
る。早期診断や病態解明には、肺内でのアスベストの<br />
種類、量、分布などを、人肺組織内で特定することが<br />
不可欠だが、今まで外科的に大きな肺組織を採取しな<br />
ければならず、簡単に調べられなかった。我々は、独<br />
立行政法人<strong>日本原子力研究開発機構</strong>(以下「原子力機<br />
構」)と 21 世紀 COE プログラムの一環として共同研<br />
究を組織した。原子力機構が開発した大気マイクロ<br />
PIXE 分析技術を応用して、数 mg の肺組織の中のケイ<br />
素や金属元素の二次元分布を、1m の解像度で画像化<br />
する分析法を開発した。本法により世界で初めて、吸<br />
入したアスベストを肺組織中に存在したままで画像化<br />
することに成功し、2008 年 10 月に International journal of<br />
immunopathology and pharmacology に論文掲載された<br />
(文献 1)。この新方式は、気管支鏡などで簡単に採取<br />
できるわずかな肺組織があれば、肺組織中のアスベス<br />
トの正確な存在や組成分析を可能にする画期的なもの<br />
である。これにより、アスベスト吸引の有無を早期に<br />
診断し、その後の迅速な対処を可能にした。その他に、<br />
環境からの粉じん暴露による肺内の重金属沈着の有無<br />
の診断など、種々の応用が期待される。病態解明では、<br />
我々は、アスベストの周囲に一致して、ヘモグロビン<br />
を貪食するマクロファージ(CD163 発現細胞)の集積と<br />
アポトーシスを引き起こし肺線維化に関与する Fas の<br />
発現が増強していることを明らかとした。しかも、<br />
Fig. 1 に示すごとく、アスベスト肺患者の肺組織に Fas<br />
や CD163 が発現している部分と発現していない部分で、<br />
アスベスト成分である Si,Fe,Mg の量を S に対する比率<br />
で求めると、明らかに Fas,CD163 発現部位でこれらの<br />
元素が高値を示しており、アスベストの存在が周囲に<br />
炎症を惹起し、これらタンパク質の発現を誘導してい<br />
ることが示唆された(文献 2)。大気マイクロ PIXE に<br />
よる研究は、吸入粒子により引き起こされた免疫反応の<br />
病態解明や吸入物質の有害レベルの決定に有用である。<br />
