In order to preparation of Biuret reagent (C), all C components in above Table were dissolved in
100 ml 0.2 M NaOH then increased volume to 250 ml with deionized water by volumetric flask
and if a black precipitate forms, it should be thrown away.
Pipette (in ml) the following reagents into suitable containers:
Reagent Test STD 1 STD 2 STD 3 STD 4 STD 5 Blank
A (NaCl) ─ 0.96 0.9 0.8 0.4 ─ 1.00
B (WPS) ─ 0.04 0.1 0.2 0.6 1.00 ─
D (Protein) 1.00 ─ ─ ─ ─ ─ ─
Mix by swirling then add (C)
C (Biuret) 4.00 4.00 4.00 4.00 4.00 4.00 4.00
Mix above samples thoroughly by vortexing and incubate for 30 minutes at 25°C. Transfer to
suitable cuvettes and record the absorbance at 540 nm for Test, Standards, and Blank. The standard
curve absorbance measurement of zygomycetes protein is shown in Figure 30 (Part 3.1.Ⅱ.).
