Abstract: Most successful vaccines currently in use are based on the generation of protective<br />
antibodies. However, CD8 + T cell responses are crucial in the <strong>de</strong>fense against several infectious<br />
agents, as well as for the treatment of cancer or chronic diseases. Thus, the <strong>de</strong>velopment of vaccine<br />
strategies capable of eliciting robust CD8 + T cell responses is absolutely nee<strong>de</strong>d. Antigen crosspresentation<br />
is known to be an important mechanism for the activation of antigen-specific CD8 + T<br />
cells, and it has been shown that multiple param<strong>et</strong>ers contribute to the efficiency of cross-priming. We<br />
examined two of them in d<strong>et</strong>ail: the route of immunization and the timing of adjuvant <strong>de</strong>livery. Our<br />
first priority was to <strong>de</strong>velop and optimize the t<strong>et</strong>ramer-based enrichment strategy, which allowed us to<br />
perform an in-<strong>de</strong>pth study of the endogenous CD8 + T cell response. This approach permitted us to<br />
work within a mo<strong>de</strong>l reflecting “physiologic” conditions in terms of initial precursor T cell frequency.<br />
We applied these m<strong>et</strong>hods to investigate the impact of the route of immunization on CD8 + T cell<br />
cross-priming. By comparing different strategies of immunization, we report that local <strong>de</strong>livery of<br />
cell-associated antigen results in <strong>de</strong>layed cross-priming due to the increased time required for antigen<br />
capture and presentation. In comparison, <strong>de</strong>livery of systemically disseminated antigen resulted in<br />
rapid T cell priming. Surprisingly, local injection of cell-associated antigen, while slower to mount a<br />
functional response, resulted in the differentiation of a more robust, polyfunctional effector T cell<br />
population and an enhanced secondary response. However, the diversity of the responding antigenspecific<br />
T cells did not appear to be affected by the route of immunization. We were next interested in<br />
evaluating the combination of cell-associated antigen with the <strong>de</strong>livery of poly I:C, an adjuvant known<br />
to induce the production of type I interferons (IFN). We observed an immunization-route-specific<br />
effect regarding the timing of innate immune stimulation and i<strong>de</strong>ntified the optimal time window for<br />
adjuvant administration in or<strong>de</strong>r to maximize the boosting effects on CD8 + T cell cross-priming. We<br />
characterized in d<strong>et</strong>ail several effects of poly I:C, as well as type I IFN, exerted on immune cells, and<br />
especially on cDCs, providing the basis for our hypotheses as to why adjuvant treatment may lead to<br />
either the inhibition or enhancement of cross-priming <strong>de</strong>pending on the timing of <strong>de</strong>livery.<br />
Résumé: La mise au point <strong>de</strong> vaccins efficaces est généralement basée sur le développement d’une<br />
réponse anticorps. Néanmoins, la protection contre certains agents infectieux ainsi que le traitement <strong>de</strong><br />
maladies chroniques ou <strong>de</strong> cancers nécessite l’induction d’une réponse cellulaire. Le développement<br />
<strong>de</strong> vaccins induisant une réponse T CD8 efficace est donc essentiel. La présentation croisée <strong>de</strong><br />
l’antigène est importante pour l’activation <strong>de</strong> lymphocytes T CD8 spécifiques. Il a été démontré que<br />
<strong>de</strong> nombreux facteurs participent au développement d’une réponse lymphocytaire T efficace. Nous<br />
nous sommes intéressés à <strong>de</strong>ux d’entre eux: la voie d’immunisation <strong>et</strong> la <strong>séquence</strong> d’aministration <strong>de</strong><br />
l’antigène <strong>et</strong> d’un adjuvant. Dans un premier temps, nous avons développé une technique<br />
d’enrichissement <strong>de</strong>s lymphocytes T CD8 spécifiques d’un antigène, ce qui a permis une étu<strong>de</strong> précise<br />
<strong>de</strong> la réponse T CD8 endogène. C<strong>et</strong>te stratégie rend possible l’analyse <strong>de</strong> la réponse lymphocytaire<br />
dans <strong>de</strong>s conditions où la fréquence initiale <strong>de</strong> précurseurs T spécifiques <strong>de</strong> l’antigène correspond aux<br />
conditions physiologiques rencontrées lors d’essais cliniques <strong>de</strong> vaccination. Nous avons utilisé c<strong>et</strong>te<br />
approche pour étudier l’influence <strong>de</strong> la voie d’immunisation sur l’efficacité <strong>de</strong> la réponse<br />
lymphocytaire T CD8. Nous avons observé que l’injection intra<strong>de</strong>rmique d’un antigène cellulaire<br />
induit une réponse T CD8 plus tardive, comparée à une administration par voie systémique.<br />
Cependant, la réponse T CD8 induite par une injection locale <strong>de</strong> l’antigène est plus efficace, avec <strong>de</strong><br />
nombreux lymphocytes capables <strong>de</strong> sécréter plusieurs cytokines. Alors que la fonctionalité <strong>de</strong>s<br />
lymphocytes T CD8 spécifiques <strong>de</strong> l’antigène dépend <strong>de</strong> la voie d’immunisation, leur diversité <strong>et</strong> leur<br />
avidité ne sont pas régulées par ce paramètre. Nous avons ensuite évalué l’administration d’un<br />
adjuvant – le poly I:C connu pour induire la production d’interférons (IFN) <strong>de</strong> type I – en parallèle <strong>de</strong><br />
celle <strong>de</strong> l’antigène. Nous avons montré que le moment optimal d’administration <strong>de</strong> l’adjuvant dépend<br />
<strong>de</strong> la voie d’immunisation. De plus, il existe une durée limitée durant laquelle l’adjuvant induit <strong>de</strong>s<br />
eff<strong>et</strong>s positifs sur l’activation <strong>de</strong>s lymphocytes T CD8. Nous avons i<strong>de</strong>ntifié plusieurs eff<strong>et</strong>s du poly<br />
I:C <strong>et</strong> <strong>de</strong>s IFN <strong>de</strong> type I sur les cellules du système immunitaire, <strong>et</strong> plus particulièrement les DCs. Ces<br />
observations nous ont permis <strong>de</strong> comprendre comment un même adjuvant pouvait avoir <strong>de</strong>s eff<strong>et</strong>s<br />
opposés en fonction du moment où il était administré.<br />
