ispitanika. Nasuprot tome, razina IL-1 alfa i IL-1 beta nije<br />
bitno povišena (0-28%). Koncentracija ostalih citokina<br />
(IL-4, IL-10, VEGF, EGF, IFN-gama) sporadično je povišena<br />
samo u nekih ispitanika (0-40%), s iznimkom IL-2 koncentracija<br />
kojega je povišena u oko 85% ispitanika. Navedeni<br />
rezultati pokazuju kako istodobno dokazivanje brojnih citokina<br />
može biti vrlo informativno, ali interpretativno vrlo<br />

Uspoređivanje testa AtheNA Multi-Lyte ANA<br />
za određivanje autoantitijela s indirektnom<br />
imunofl uorescencijom i testom ELISA<br />

Laboratorijski testovi koji se rabe u obradi bolesnika s autoimunim<br />
bolestima služe za dijagnosticiranje bolesti, kliničko<br />
praćenje bolesnika i procjenu uspješnosti liječenja.<br />
Test AtheNA Multi-Lyte ANA rabi metodu homogene fl uoroimunokemije<br />
gdje su polistirenske kuglice obložene s<br />
više različitih antigena. Stoga ovaj test služi za polukvantitativno<br />
dokazivanje IgG klase antitijela za SS-A, SS-B, Sm,<br />
RNP, Scl-70, Jo-1, centromera i histona, te za kvantitativno<br />
dokazivanje IgG klase antitijela na dsDNA i kvalitativno<br />
dokazivanje IgG klase antitijela na ANA. Cilj rada je bio usporediti<br />
test AtheNA Multi-Lyte ANA s konvencionalnim<br />
metodama indirektne imunofl uorescencije (IIF) i enzimimunoanalize<br />
(ELISA) koje rabimo u našem svakodnevnom<br />
radu. Analizirali smo 152 uzorka seruma zaprimljenih u laboratorij<br />
sa zahtjevima za određivanje slijedećih antitijela:<br />
97 ANA, 59 SS-A, 56 SS-B, 11 Sm, 9 Sm/RNP, 39 Scl-70, 17<br />
Jo-1, 39 dsDNA, 16 histona i 19 centromera bez obzira na<br />
dijagnozu. Svi uzorci usporedno su određivani i mjereni<br />
testom AtheNA Multi-Lyte ANA (Zeus Scientifi c, Inc.) na<br />
instrumentu Luminex 100 IS, komercijalnim testom ANA<br />
Hep-2 (BioSystems) na fl uorescentnom mikroskopu, komercijalnim<br />
testom ELISA za dokazivanje ENA antitijela<br />
(SS-A, SS-B, Sm, Sm/RNP, Scl-70, Jo-1), dsDNA (Hycor test)<br />
te histona i centromera (DiaSorin) na automatskom analizatoru<br />

Elevated concentrations of IL-6 and TNF-alfa were found<br />
in 75% of infants and adults. At the same time, the concentrations<br />
of IL-1 alfa and IL-1 beta were not signifi cantly<br />
elevated (0-28%). Plasma levels of other cytokines (IL-4,<br />
IL-10, VEGF, EGF, IFN-gamma) were only sporadically elevated<br />
(0-40%), with the exception of IL-2. Plasma levels<br />
of this cytokine were elevated in 85% of patients. These<br />
data suggest that simultaneous determination of multiple<br />
proinfl ammatory cytokines and growth factors by<br />
biochip array may be very informative but very complex<br />
in terms of interpretation.<br />

Measurement of autoantibodies provides supporting<br />
evidence in the diagnosis and monitoring of systemic<br />
autoimmune diseases. The AtheNA Multi-Lyte ANA Test<br />
System (Zeus Scientifi c, Inc.) is a homogeneous, multiplexed,<br />
fl uorescence-based microparticle immunoassay<br />
intended for semi-quantitative detection of IgG class<br />
antibody to 8 separate analytes (SS-A, SS-B, Sm, RNP,<br />
Scl-70, Jo-1, centromere B and histone), quantitative detection<br />
of IgG class antibody to dsDNA, and qualitative<br />
detection of IgG class antibody to ANA in human serum.<br />
The objective of this study was to determine the performance<br />
of the AtheNA Multi-Lyte ANA test relative to the<br />
established, commercial indirect immunofl uorescence<br />
(IIF) and enzyme-linked immunosorbent assay (ELISA)<br />
currently in use in our laboratory. The study population<br />
consisted of 152 serum specimens referred to our laboratory<br />
for autiommune testing: 97 ANA requests, 59 SS-A,<br />
56 SS-B, 11 Sm, 9 Sm/RNP, 39 Scl-70, 17 Jo-1, 39 dsDNA, 16<br />
histone and 19 centromere. Diagnosis was not included<br />
in the study. All specimens were tested on the AtheNA<br />
Multi-Lyte ANA Test System and a multitude of conventional<br />
immunoassays including commercial ANA Hep-2<br />
test systems (BioSystems), commercial ELISA test systems<br />
for detection of antibodies to ENA (SS-A, SS-B, Sm, Sm/<br />
