The Bureau <strong>of</strong> Citrus <strong>Budwood</strong> Registration oversees budwood<br />

certification in <strong>Florida</strong>’s commercial citrus nurseries. The protection <strong>of</strong><br />

<strong>Florida</strong>’s propagating material is fundamental to ensuring growers<br />

receive disease-free trees to plant. Citrus growers have enough<br />

challenges without having to worry if the nursery stock they are planting<br />

is healthy. Planting pathogen-infected trees is economically<br />

unsustainable <strong>and</strong>, at the very least, farm pr<strong>of</strong>its would be greatly<br />

diminished. Disease-infected trees <strong>of</strong>ten are not noticed until after<br />

growers have absorbed several years <strong>of</strong> caretaking expenses on top <strong>of</strong><br />

tree <strong>and</strong> planting costs. <strong>Budwood</strong> registration includes grafttransmissible<br />

pathogen testing <strong>of</strong> all propagating sources in <strong>Florida</strong>.<br />

Allowing only disease-tested material into the nurseries increases the<br />

likelihood <strong>of</strong> disease-free plants leaving the nursery. The Bureau <strong>of</strong><br />

Citrus <strong>Budwood</strong> Registration’s foundation facility provides nurserymen<br />

with tested scion material for establishment <strong>of</strong> their own clean stock. The<br />

bureau’s laboratory tests foundation <strong>and</strong> nursery propagating source<br />

routinely for the endemic graft-transmissible diseases <strong>of</strong> citrus.<br />

This year’s high point was the completion <strong>of</strong> the Chiefl<strong>and</strong> budwood<br />

foundation facility. The rapid start-up <strong>of</strong> this project was a result <strong>of</strong><br />

having clean, protected trees available to move from a central <strong>Florida</strong><br />

greenhouse to the new greenhouse - without having to start over with<br />

newly budded trees. The fact that those pathogen-tested trees had been<br />

under protected greenhouse cover their entire lives, accelerated the<br />

transition. Therefore, budwood was made available almost from day one.<br />

The Chiefl<strong>and</strong> greenhouses are in their second year <strong>of</strong> operation, <strong>and</strong> are<br />

starting to produce significant quantities <strong>of</strong> budwood. One hundred<br />

thirty-two thous<strong>and</strong> budeyes were cut from the Chiefl<strong>and</strong> greenhouses<br />

this fiscal year, bringing the total budwood cut in the first 1½ years to<br />

174,210 eyes.<br />

Nine hundred seventy-one tree spaces are planted at Chiefl<strong>and</strong>, leaving<br />

room for only 164 additional trees. The planted trees represent 321<br />

different clonal selections. The Chiefl<strong>and</strong> foundation planting is 33.9%<br />

sweet orange, representing 52 different sweet orange varieties. M<strong>and</strong>arins<br />

make up 27.0% <strong>of</strong> the trees, representing 73 varieties. Grapefruit is<br />

represented with 22 varieties <strong>and</strong> consists <strong>of</strong> 9.3% <strong>of</strong> the planting. The<br />

greenhouse trees are inspected every day. Also, the perimeter <strong>of</strong> the<br />

structure is walked daily, being closely scanned for any possible areas<br />

that could allow insect incursion.<br />

The lowest temperature recorded outside the greenhouse during the<br />

winter <strong>of</strong> 2009 was 17 degrees. Inside the greenhouses the temperature<br />

A new <strong>of</strong>fice/headhouse building was added to the bureau’s Chiefl<strong>and</strong><br />

foundation facility in 2008-2009. The new <strong>of</strong>fice includes a walk-in cooler<br />

for the most favorable storage <strong>of</strong> budwood before shipment. A large work-<br />

room allows for trimming budwood in an air-conditioned environment,<br />

further optimizing the condition <strong>of</strong> the budwood. A conference room<br />

provides a place for small groups to meet <strong>and</strong> receive information on the<br />

budwood facility without having to enter the greenhouses. Because <strong>of</strong> the<br />

importance <strong>of</strong> minimizing any risk <strong>of</strong> pathogen contamination <strong>of</strong> the<br />

foundation clean stock, the greenhouse areas are <strong>of</strong>f limits to visitors.<br />

Although a window has been placed in the side <strong>of</strong> the greenhouse for visitors<br />

to view the operation.<br />

The Immokalee IFAS cooperative foundation greenhouse facility, which has<br />

not been used for budwood since the 2006-07 fiscal year, has been partially<br />

top-worked to seed source varieties. Eleven different seed source varieties<br />

have been top-worked inside <strong>of</strong> greenhouse #2.<br />

Since the advent <strong>of</strong> <strong>Florida</strong>’s budwood program, more than 163 million<br />

registered nursery trees have been produced. Nursery capacity <strong>and</strong><br />

production has been increasing the past three years, after a four year period <strong>of</strong><br />

decline. Production levels have nearly returned to what was produced in the<br />

2003-2004 season. Although the number <strong>of</strong> citrus nurseries is lower than we<br />

had five years ago, the average production per nursery has increased. The<br />

average nursery made 85 thous<strong>and</strong> propagations this fiscal year. Forty-six<br />

percent <strong>of</strong> <strong>Florida</strong>’s nursery trees are produced in Polk County, <strong>and</strong> this is<br />

where the majority <strong>of</strong> the citrus nursery inspectors are stationed. Forty-four<br />

commercial citrus nurseries reported making nearly 3.8 million propagations<br />

in 15 counties.<br />

The bureau registered 8,665 scion trees this fiscal year. This is up 594 trees<br />

from last year. This represents 5,515 trees (64%) that are budwood sources<br />

<strong>and</strong> 3,150 trees (36%) that are seed source trees. <strong>Budwood</strong> sources are<br />

required to be grown in protected greenhouses that have to be approved by<br />

the Division <strong>of</strong> Plant Industry, while seed source trees can be field grown.<br />

Bureau personnel witnessed the planting <strong>of</strong> 933 new scion trees this fiscal<br />

year, <strong>of</strong> which 415 are seed source. The average age <strong>of</strong> a budwood scion tree<br />

is 3.2 years old. The transformation <strong>of</strong> the nursery industry from field to<br />

greenhouse in 2006 resulted in newly established protected budwood trees.<br />

While initially reducing the availability <strong>of</strong> budwood, the longer term result is<br />

cleaner pathogen-free propagation material. Scion trees produced 51% <strong>of</strong> the<br />

nursery propagating material, with<br />

the average scion tree producing 878<br />

budeyes in 2008-2009. Increase<br />

trees accounted for 45% <strong>of</strong> nursery<br />

propagation material. Qualifying<br />

increase trees, those originating<br />

from foundation trees, can be<br />

converted to scion trees by being<br />

witnessed <strong>and</strong> pathogen tested.<br />

This fiscal year, all program seed source <strong>and</strong> budwood scion trees were tested<br />

for citrus greening with no greening-positive trees being found. Seed source<br />

trees were all additionally tested for citrus leaf blotch virus, with only 2 trees<br />

testing positive. <strong>Budwood</strong> scion trees were also tested for citrus tristeza virus<br />

<strong>and</strong> citrus viroids. Collecting the samples from all 8,665 scion trees takes over<br />

3 months. The chopping <strong>of</strong> the samples is done immediately upon arrival in the<br />

lab. Testing begins with all trees being run for citrus greening, before any other<br />

tests are conducted. Citrus tristeza virus testing begins once all the HLB tests<br />

have been completed. Additional pathogen testing then begins, once the tristeza<br />

tests are completed. All these tests are qPCR based, <strong>and</strong> the initial extraction is<br />

suitable for all subsequent tests. This is a very efficient <strong>and</strong> cost- effective<br />

means <strong>of</strong> testing source trees, as only<br />

one collection per year is required.<br />

In past years, multiple trips to nursery<br />

sites were required for sample<br />

collection. One trip is all that is now<br />

needed, due to the ability to detect all<br />

pathogens from the one extraction. A<br />

total <strong>of</strong> 33,118 pathogen tests were run<br />

at the bureau’s Winter Haven laboratory. The majority <strong>of</strong> these analyses were<br />

on scion trees, <strong>and</strong> foundation trees. The <strong>Florida</strong> Citrus Arboretum had 2<br />

additional HLB positive trees this year, bringing the total <strong>of</strong> positives at this<br />

location to 4. No program budwood source trees were identified as being<br />

greening infected. The old foundation grove at Dundee continues to struggle<br />

with citrus canker infection. Approximately 644 grapefruit trees were removed<br />

from the grove to reduce the risk <strong>of</strong> spreading canker inoculum to the sweet<br />

oranges. The original seed source block containing many Poncirus trifoliate<br />

hybrids, has been particularly hard hit by canker infection, with 65 percent <strong>of</strong><br />

the trees being removed.<br />

Hamlins <strong>and</strong> Valencias, the perennial favorite varieties produced, have<br />

consistently been the top two citrus varieties grown. Midsweet continues in<br />

third place, <strong>and</strong> is the top midseason variety. With <strong>Florida</strong>’s citrus industry<br />

predominately producing an orange juice product, it is no surprise that these<br />

three sweet varieties are the most widely planted. Sweet oranges comprise 89<br />

percent <strong>of</strong> the total nursery production. Grapefruit represent 4.7 % <strong>of</strong> nursery<br />

propagations, <strong>and</strong> m<strong>and</strong>arin 4.1%. Ray Ruby was the most widely used<br />

grapefruit variety, <strong>and</strong> is also the #1 variety being topworked. Ruby Red is<br />

the second top grapefruit selection propagated, followed by Rio Red.<br />

year. Although the citrus industry<br />

utilizes only a minority <strong>of</strong> varieties in<br />

commercial plantings, the dooryard<br />

(homeowner) market employs a vast<br />

array <strong>of</strong> citrus types. Swingle<br />

citrumelo remains the top used<br />

rootstock, a position it has held now for 21 years. Nursery trees on Swingle<br />

account for 40 % <strong>of</strong> all propagations. Kuharske citrange <strong>and</strong> Carrizo citrange<br />

were the 2 nd <strong>and</strong> 3 rd most utilized rootstocks, accounting for 24% <strong>and</strong> 18% <strong>of</strong><br />

<strong>Budwood</strong> Program<br />

The average nursery produced 85,551 trees in 2009 Topworkers 3<br />

Over 163 million registered trees have been propagated since 1953<br />

46 % <strong>of</strong> nursery trees are produced in Polk county<br />

Participating <strong>Florida</strong> Citrus Nurseries<br />

Many commercial nurseries also sell dooryard trees <strong>and</strong> several<br />

are mainly own use, but keep the option to sell to other growers<br />

as well.<br />

Forty-four active <strong>Florida</strong> citrus nurseries reported making propagations in the 2008– 2009 fiscal year.<br />

<strong>Florida</strong> nursery propagations increased 6% from the previous fiscal year.<br />

Historical look at the numbers <strong>of</strong> tests run for routine citrus pathogen indexing. A larger number <strong>of</strong> indicator plants were used as the number <strong>of</strong> indicators<br />

per test varied from 3 to 10.<br />

Pathogen Testing<br />

Testing for Citrus greening (HLB) <strong>and</strong> Citrus tristeza virus (CTV) are done annually as both <strong>of</strong> these diseases<br />

have insect vectors. This test ensures that if any insects were able to breach the greenhouse approved structures<br />

<strong>and</strong> transmit either <strong>of</strong> these diseases it would be detected. There is one annual collection <strong>of</strong> samples <strong>and</strong> it starts<br />

in late August <strong>and</strong> continues until all collections are complete with December as the goal for completion. As<br />

HLB is our most important pathogen, this time <strong>of</strong> year for collection was chosen as it is the optimal time <strong>of</strong> year<br />

to scout for HLB <strong>and</strong> also for its detection. As we do not have a dedicated collection crew, Bureau laboratory,<br />

grove <strong>and</strong> inspection personnel from the Winter Haven <strong>of</strong>fice collect the majority <strong>of</strong> the samples. Staff at the<br />

Chiefl<strong>and</strong> Foundation help with the collections from the northern nurseries in their vicinity. This fiscal year our<br />

laboratory extracted 10,265 samples. Utilizing these extractions 7,128 scion trees <strong>and</strong> 926 Chiefl<strong>and</strong> trees were<br />

tested for HLB using a real time PCR (qPCR test). From the same extract used for the HLB test, CTV qPCR<br />

tests were run also. CTV qPCR tests have replaced the ELISA tests that required a separate spring collection.<br />

These qPCR tests are more sensitive than the MCA-13 ELISA test, use the same principle <strong>and</strong> can distinguish<br />

MCA-13 severe strains from mild strains <strong>of</strong> CTV.<br />

Citrus leaf blotch virus (CLBV) is an important virus as we have found it in some <strong>of</strong> the source trees <strong>and</strong> it can<br />

be seed transmitted. We developed a qPCR test that we used this past year to test all seed source trees as well as<br />

scion <strong>and</strong> foundation trees. A low level <strong>of</strong> positive trees were detected <strong>and</strong> removed. Real-time PCR testing for<br />

three diseases using the same extract from one collection has allowed sensitive detection <strong>and</strong> maximized the<br />

time <strong>and</strong> effort used for the collections from over 10,000 trees.<br />

Shoot-Tip Grafting <strong>and</strong> <strong>Florida</strong> Parent Tree Introduction<br />

The application <strong>of</strong> the efficient qPCR testing technology to incoming parent c<strong>and</strong>idate trees <strong>and</strong> their shoot-tip<br />

grafts (STGs) has helped to decrease the length <strong>of</strong> time for testing. Tests on incoming parent c<strong>and</strong>idates give us<br />

an early indication <strong>of</strong> what disease(s) need(s) to be eliminated. Because <strong>of</strong> the potential <strong>of</strong> Citrus greening in<br />

incoming parent tree introductions, they are automatically shoot-tip grafted to remove this risk. QPCR tests on<br />

STGs allow early determination <strong>of</strong> disease status while slower biological tests are in progress. After initial<br />

qPCR testing results are obtained, a horticulturally superior specimen is selected, grown <strong>of</strong>f <strong>and</strong> increased for<br />

eventual release.<br />

We recognize the importance <strong>of</strong> having all possible <strong>Florida</strong> Germplasm selections as disease free as possible<br />

<strong>and</strong> kept safe in our new screen house facility at Chiefl<strong>and</strong>. This fiscal year an emphasis was placed on the<br />

Parent Tree Indexing <strong>and</strong> Shoot-tip grafting programs to expedite progress on all selections <strong>and</strong> also to work to<br />

remove mild isolates <strong>of</strong> CTV from clones at Chiefl<strong>and</strong> where necessary.<br />

Sixty-two trees were in the Parent Tree Indexing Program at year’s end. Twenty-five were entered this year <strong>and</strong><br />

32 parents <strong>and</strong>/or STGs were released this year. Of the released clones, five were Breeder’s varieties, eight were<br />

already available at Chiefl<strong>and</strong>, but had the Mild CTV removed, nine were STGed to remove severe CTV, three<br />

viroids, <strong>and</strong> one Citrus leaf blotch virus.<br />

In 2008-2009, we had our greatest production <strong>of</strong> shoot-tip grafts, doubling that <strong>of</strong> the previous year. 1,846<br />

shoot-tip grafts were set up, <strong>of</strong> these, 135 were grafted into rootstocks in the greenhouse. At the end <strong>of</strong> the fiscal<br />

year a total <strong>of</strong> 43 cultivars were in the process <strong>of</strong> being shoot-tip grafted. Fifty-six cultivars have been<br />

successfully STGed <strong>and</strong> are undergoing various stages <strong>of</strong> testing. The process <strong>of</strong> STGing a selection used to<br />

take a minimum <strong>of</strong> five years. Now for selections newly entered into the program, the entire process takes under<br />

two years if there are no complications.<br />

Our Vision<br />

The Bureau <strong>of</strong> Citrus <strong>Budwood</strong> Registration will provide high yielding, pathogen free, quality budlines that will<br />

positively impact the productivity <strong>and</strong> prosperity <strong>of</strong> our citrus industry.<br />

Our Mission<br />

The Bureau <strong>of</strong> Citrus <strong>Budwood</strong> Registration administers a program to assist growers <strong>and</strong> nurserymen in<br />

producing citrus nursery trees that are believed to be horticulturally true to varietal type, productive, <strong>and</strong> free<br />

from certain recognizable bud-transmissible diseases detrimental to fruit production <strong>and</strong> tree longevity.<br />

Benefits <strong>of</strong> a <strong>Budwood</strong> Program<br />

The <strong>Florida</strong> budwood program has greatly impacted <strong>Florida</strong> citrus, as the use <strong>of</strong> registered budwood selections<br />

or “budlines” is responsible for impressive increases in yield <strong>and</strong> fruit quality in the industry. The budwood<br />

program introduced many nucellar lines that are still the basis <strong>of</strong> many <strong>of</strong> our main varieties. The ability to test<br />

for graft-transmitted pathogens <strong>and</strong> exclude unclean propagating material has not only increased production <strong>and</strong><br />

tree longevity but also the ability <strong>of</strong> our groves to withst<strong>and</strong> other environmental stresses.<br />

• Diseases are kept out <strong>of</strong> the propagation system<br />

o Using clean nursery stock means growers receive disease free trees coming out <strong>of</strong> the nursery<br />

• Testing for citrus viroids is more critical than ever as the industry uses predominantly citrange<br />

<strong>and</strong> citrummelo type rootstocks that are more susceptible to viroids<br />

o The budwood program is credited with having viroid free trees for the industry to use when the rootstocks<br />

were changed from rough lemon <strong>and</strong> sour orange to Carrizo <strong>and</strong> Swingle<br />

Other countries without budwood programs had major viroid problems when changing rootstocks<br />

away from sour orange<br />

• Citrus tristeza virus strains are worse today than a few years ago<br />

o Stem pitting tristeza virus strains have been found in several commercial grove locations in <strong>Florida</strong><br />

• Psorosis virus is becoming hard to find in <strong>Florida</strong> after 50 years <strong>of</strong> budwood registration<br />

o Prior to the budwood program psorosis was widespread in <strong>Florida</strong><br />

Yields in psorosis infected trees were reduced 10 to 35 percent <strong>and</strong> made the trees easier to<br />

succumb to cold <strong>and</strong> other environmental stresses<br />

• Protection <strong>of</strong> our industry from exotic citrus diseases requires controlling citrus propagating<br />

material<br />

o Citrus greening is the latest disease to move from the exotic disease list to become an endemic disease<br />

o Citrus variegated chlorosis <strong>and</strong> citrus sudden death are examples <strong>of</strong> pathogens whose main means <strong>of</strong><br />

control is budwood certification<br />

Grove trees started from clean stock have the potential to be pr<strong>of</strong>itable even in the presence <strong>of</strong><br />

disease, whereas nursery trees started systemically infected will never become pr<strong>of</strong>itable grove<br />

trees<br />

• Yields are increased with registered budwood clonal selections<br />

o Nucellar Valencia clones alone increased yields by 25% over old line selections<br />

o Examples <strong>of</strong> registered clonal selections in every variety show remarkable yield gains<br />

• True to type varieties <strong>and</strong> rootstocks are propagated<br />

o Inspected trees with <strong>of</strong>f-types are removed from budwood supply<br />

• <strong>Budwood</strong> programs are recognized world wide as the most effective means <strong>of</strong> excluding <strong>and</strong><br />

containing citrus graft-transmissible pathogens<br />
