Description: DNA Polymerase I Large (Klenow) Fragment, Exonuclease Minus,<br />

is a DNA-dependent DNA polymerase that lacks both the 5´→3´ and the 3´→5´<br />

exonuclease activities present in intact E. coli DNA Polymerase I. It is used for<br />

random primer labeling and in strand displacement amplification. Klenow<br />

Fragment, Exonuclease Minus, will leave a single-base 3´ overhang on a<br />

significant proportion of DNA fragments during fill-in of 5´-overhangs.<br />

Therefore, this enzyme is not recommended for preparation of blunt-ended<br />

fragments for ligation.<br />

Features:<br />

• Provided with 10X Reaction Buffer: 500mM Tris-HCl (pH 7.2 at 25°C),<br />

100mM MgSO 4, 1mM DTT.<br />

• May Be Heat-Inactivated: DNA Polymerase I Large (Klenow) Fragment,<br />

Exonuclease Minus, is inactivated by heating at 75°C for 10 minutes.<br />

Description: T4 DNA Polymerase catalyzes the 5´→3´ synthesis of DNA from<br />

a primed single-stranded DNA template. Although possessing a potent 3´→5´<br />

proofreading exonuclease, T4 DNA Polymerase contains no 5´→3´ exonuclease<br />

activity. T4 DNA Polymerase can be used to fill 5´ protruding ends with labeled<br />

or unlabeled dNTPs or for the generation of blunt ends from DNA molecules<br />

with 3´ overhangs.<br />

• High Fidelity: T4 DNA Polymerase is the enzyme of choice for applications<br />

where misincorporation is a concern.<br />

• Flexible: T4 DNA Polymerase may be used in a variety of molecular<br />
