Description: The CytoTox 96 ® Non-Radioactive Cytotoxicity Assay is a<br />

colorimetric alternative to radioactive cytotoxicity assays. The CytoTox 96 ®<br />

Assay quantitatively measures lactate dehydrogenase (LDH), a stable cytosolic<br />

enzyme that is released upon cell lysis, in much the same way as [ 51 Cr] is<br />

released in radioactive assays. Released LDH in cell culture supernatants<br />

is measured with a 30-minute coupled enzymatic assay that results in the<br />

conversion of a tetrazolium salt (INT) into a red formazan product. The amount<br />

of color formed is proportional to the number of lysed cells. Visible wavelength<br />

absorbance data are collected using a standard 96-well plate reader. The assay<br />

can be used to measure membrane integrity for cell-mediated cytotoxicity<br />

assays in which a target cell is lysed by an effector cell, or to measure lysis of<br />

target cells by bacteria, viruses, proteins, chemicals, etc.<br />

Description: The Griess Reagent System measures nitrite (NO 2 – ), which<br />

is one of two primary stable and nonvolatile breakdown products of nitric<br />

oxide (NO). Nitric oxide is an important physiological messenger and effector<br />

molecule in many biological systems, including immunological, neuronal and<br />

cardiovascular tissues. This assay relies on a diazotization reaction that was<br />

originally described by Griess in 1879. Through the years, many modifications<br />

to the original reaction have been described.<br />

The Griess Reagent System is based on a chemical reaction that uses sulfanilamide<br />

and N-1-naphthylethylenediamine dihydrochloride (NED) under acidic<br />

(phosphoric acid) conditions. This system detects NO 2 – in a variety of biological<br />

and experimental liquid matrices such as plasma, serum, urine and tissue<br />

culture medium. The nitrite sensitivity is dependent on the matrix. The limit of<br />

detection is 2.5μM (125pmol) nitrite (in ultrapure, deionized, distilled water)<br />

using the protocol described in Technical Bulletin #TB229.<br />

Storage Conditions: Store at 4°C. Keep all solutions in their original lightprotective<br />

plastic bottles.<br />
