Description: The RiboMAX Large Scale RNA Production Systems produce<br />
2–5mg/ml of RNA in a 1ml reaction, about 10- to 20-fold more RNA than is<br />
produced with the standard Riboprobe ® System transcription reaction. The<br />
RiboMAX System reactions differ from those of the Riboprobe ® Systems in<br />
three primary ways: a HEPES (pH 7.5) buffer is used rather than a Tris-HCl<br />
(pH 7.9) buffer; rNTP and magnesium concentrations are elevated at levels<br />
appropriate for either SP6 or T7 RNA polymerase; and inorganic<br />
pyrophosphatase is included in the reaction.<br />
RNAs synthesized with the RiboMAX System perform better for in vitro<br />
translation in rabbit reticulocyte translation systems than RNA synthesized by<br />
standard methods. The reduction of components inhibitory to translation may be<br />
advantageous for other applications requiring biologically active RNA. Because<br />
the RiboMAX Systems produce large quantities of RNA, these systems are not<br />
recommended for the generation of high-specific-activity RNA probes.<br />

Description: The T7 RiboMAX Express Large Scale RNA Production System<br />
is an in vitro transcription system designed for the consistent production of<br />
milligram amounts of RNA in a short amount of time. Due to optimization of the<br />
enzyme mix and transcription buffer, yields of 5–8.5mg/ml are generated in 30<br />
minutes, compared to 2–4 hours with other commercially available systems. To<br />
minimize pipetting steps and errors, the 2X transcription buffer includes all four<br />
rNTPs. In addition, the system includes RQ1 RNase-Free DNase for the removal<br />
of plasmid template after transcription.<br />
Due to the combined 2X buffer and rNTPs, the T7 RiboMAX Express<br />
System is not recommended for the synthesis of RNA for applications that<br />
require capped RNA. For synthesis of capped RNA, please order the standard<br />
RiboMAX Large Scale RNA Production System—T7 (Cat.# P1300).<br />

Description: The Riboprobe ® Systems are designed for in vitro preparation<br />
of high-specific-activity single-stranded RNA probes or microgram quantities<br />
of defined RNA transcripts from cloned DNA inserts. These systems contain all<br />
components necessary for in vitro transcription from a DNA template (excluding<br />
the radioisotope) and also contain RQ1 RNase-Free DNase (Cat.# M6101) for<br />
template removal following transcription.<br />
