Description: ViviRen Live Cell Substrate is a uniquely engineered<br />

coelenterazine that generates three- to fivefold brighter Renilla luciferase<br />

luminescence than wildtype coelenterazine. Using live cells, achieve up to<br />

100-fold higher signal-to-noise ratios for super-sensitive quantitation of<br />

reporter gene, BRET and RNAi activity.<br />

Description: Luciferases from Renilla, Aequorea and other marine organisms<br />

are commonly used as indicators or reporters for studying cellular phenomena<br />

in expression assays in eukaryotic cells. Renilla luciferase is often used as a<br />

reporter of transcription regulation, whereas apoaequorin is often used as a<br />

calcium indicator. Other uses of coelenterazines include chemiluminescent<br />

detection of Reactive Oxygen Species (ROS) in cells or tissues. <strong>Promega</strong> offers<br />

the following coelenterazine analogs.<br />

Coelenterazine (native) is the luminescent substrate for Renilla luciferase<br />

and apoaequorin. Formula: C 26H 21N 3O 3. Formula Weight: 423.5. Form: Film.<br />

Coelenterazine-h imparts a luminescent intensity with its aequorin complex<br />

that is reported to be 10–20 times higher than that of native coelenterazine,<br />

making this derivative a useful tool for measuring small changes in Ca 2+<br />

Description: The β-Galactosidase Enzyme Assay System with Reporter Lysis<br />

Buffer is a convenient method for assaying β-galactosidase activity in lysates<br />

prepared from cells transfected with β-galactosidase reporter vectors such as<br />

the pSV-β-Galactosidase Control Vector.<br />

The standard assay is performed by adding a dilute sample to an equal<br />

volume of Assay 2X Buffer that contains the substrate ONPG (o-nitrophenylβ-d-galactopyranoside).<br />

Samples are incubated for at least 30 minutes,<br />

during which time the β-Galactosidase hydrolyzes the colorless substrate to<br />

o-nitrophenyl, which is yellow. The reaction may be terminated by addition of<br />

sodium carbonate, and the absorbance at 420nm is measured by<br />

spectrophotometry.<br />
