Description: The purified Anti-VAChT (Vesicular Acetylcholine Transporter) pAb<br />

is raised in goats against a peptide (CSPPGPFDGCEDDYNYYSRS) corresponding<br />

to amino acids 511–530 of the carboxy terminus of the cloned rat VAChT. It is a<br />

novel tool to identify functional cholinergic neurons in the central and peripheral<br />

nervous system where the antibody stains fibers and neuronal cell bodies. This<br />

antibody has application for the study of the pathophysiology of neurodegenerative<br />

diseases involving the cholinergic system and for mapping cholinergic<br />

neurons in the nervous system.<br />

Features:<br />

• Immunogen: Carboxy-terminal peptide sequence 511–530 corresponding<br />

to cloned rat VAChT protein.<br />

• Antibody Form: Purified goat polyclonal IgG; 0.5mg/ml in PBS containing<br />

no preservatives.<br />

• Specificity: Cross-reacts with VAChT in rat and mouse, but not in human,<br />

guinea pig, rabbit or cat.<br />

Immunofluorescence staining for VAChT protein in ventral gray matter<br />

of rat spinal cord. VAChT was detected using Anti-VAChT pAb at<br />

1μg/ml followed by anti-goat Cy ® 3 conjugate. Specificity was demonstrated by<br />

preabsorption of Anti-VAChT pAb (1μg/ml) with the peptide immunogen<br />

(5μg/ml) prior to incubation with the primary antibody and anti-goat Cy ® 3<br />

conjugate (data not shown). Protocols developed and performed at <strong>Promega</strong>.<br />

Description: Donkey Anti-Rabbit Ig, (H+L), HRP, are horseradish<br />

peroxidase-conjugated secondary antibodies specifically tested for use with<br />

the Anti-ACTIVE ® antibodies. They are qualified for use in Western blot analysis<br />

using chemiluminescent and colorimetric detection methods. These antibody<br />

conjugates exhibit minimal cross-reactivity to goat, mouse and sheep IgG,<br />

bovine serum albumin (BSA) and proteins in mammalian cell extracts. These<br />

secondary antibody conjugates provide low background and highly specific<br />

signals when used at the recommended dilutions with Anti-ACTIVE ® MAPK,<br />

Anti-ACTIVE ® JNK and Anti-ACTIVE ® p38 pAbs. The conjugates are provided<br />

in phosphate-buffered saline containing BSA as a stabilizer and gentamicin as<br />

Features:<br />

• Sensitivity: When conjugates are used in a Western blot at a 1:10,000<br />

dilution along with Anti-ACTIVE ® MAPK pAb (Cat.# V8031), they can detect<br />

active MAP kinase in 5μg of activated (nerve growth factor [NGF]-treated)<br />

PC12 cell extract using colorimetric detection.<br />

• Specificity: Preferentially detects rabbit IgG with minimum reactivity with<br />

immunoglobulins from other species (including goat, sheep and mouse) or<br />

with bovine serum albumin and mammalian cell extract proteins.<br />

• Value: 60μl per vial, sufficient to generate 300–600ml of Western blotting<br />

Description: The Alkaline Phosphatase-Conjugated Antibodies are used in the<br />

detection of proteins in Western blotting and ELISAs. The alkaline phosphatase<br />

(AP) catalyzes colorimetric reactions using Western Blue ® Substrate (standard<br />

BCIP/NBT) or AttoPhos ® Reagent. It can also drive the chemiluminescent<br />

detection reactions involving substrates such as 3-(2´-spiroadamantane)-<br />

4-methyl-4-(3´´ phosphoryloxyphenyl-1, 2-dioxetane (AMPPD ® ).<br />

Goat Anti-Mouse IgG (H+L), Alkaline Phosphatase Conjugate is an<br />

affinity-purified goat anti-mouse antibody. It reacts with mouse IgG (all<br />

Antibodies may cross-react with immunoglobulins from other species.<br />

Goat Anti-Rabbit IgG (Fc), Alkaline Phosphatase Conjugate is an affinitypurified<br />

goat anti-rabbit antibody. It reacts with the heavy chains of rabbit IgG<br />

but not with light chains. Antibodies may cross-react with immunoglobulins<br />

from other species.<br />

Goat Anti-Human IgG (H+L), Alkaline Phosphatase Conjugate is an<br />

affinity-purified goat anti-human antibody. This product has been isolated from<br />

antisera by immunoaffinity chromatography using immobilized antigens. It<br />

reacts with all subclasses of human IgG as well as with light chains on other<br />

human immunoglobulins. Antibodies may cross-react with immunoglobulins<br />

from other species. This product displays minimal cross-reactivity with horse<br />
