Description: Anti-ACTIVE ® p38 pAb is a polyclonal rabbit antibody. The<br />

antibody is affinity-purified using a dually phosphorylated peptide that<br />

corresponds to the active form of the p38 enzymes.<br />

Features:<br />

• Specificity: Preferentially detects the dually phosphorylated, active form of<br />

p38 kinase.<br />

• Immunogen: Dually phosphorylated Thr/Gly/Tyr region (pTGpY) derived<br />

from the catalytic core of the active form of p38 kinase, which corresponds<br />

to Thr 180 and Tyr 182 of the mammalian p38 enzyme.<br />

• Antibody Form: Affinity-purified rabbit IgG; supplied in PBS (pH 7.4).<br />

• Value: When used at the recommended 1:2,000 dilution, this product will<br />

generate up to 200ml of blotting solution, sufficient for 20 Western blots of<br />

10ml each.<br />

Detection of activated MAPK, JNK and p38 in PC12 cells by<br />

immunocytochemistry. PC12 cells were grown to 80% confluence in<br />

RPMI 1640 medium supplemented with 25mM HEPES, 300mg/L l-glutamine,<br />

10% horse serum, 5% fetal bovine serum and 0.5mM EGTA. Cells were either<br />

untreated or treated with 200ng/ml NGF or 1M sorbitol as indicated. ICC was<br />

performed as described in <strong>Promega</strong> Technical Bulletin #TB262. Anti-ACTIVE ®<br />

antibodies were used at the following dilutions: Panel A. MAPK, 1:500;<br />

Panel B. JNK, 1:1,000; Panel C. p38, 1:500. Protocols developed and<br />

performed at <strong>Promega</strong>.<br />
