Description: The Block & Sample 5X Buffer is optimized for use in the E max ®<br />

ImmunoAssay Systems (for BDNF, GDNF and NGF) providing additional buffer<br />

for further sample dilutions and manipulations. This buffer is used to block the<br />

plates and dilute the standards, samples, detection antibodies and conjugates<br />

in these E max ® ImmunoAssay Systems. The buffer is provided as 54ml of buffer<br />

containing gentamicin as a preservative.<br />

Description: The TGFβ 1 E max ® ImmunoAssay System provides optimized<br />

reagents and a protocol for the sensitive and specific detection of transforming<br />

growth factor β 1 (TGFβ 1). After an overnight coating of a 96-well plate, the<br />

specific protein is detected using an antibody sandwich format. The system<br />

uses horseradish peroxidase-conjugated secondary antibody and a singlecomponent<br />

TMB substrate for the final chromogenic detection of bound TGFβ 1.<br />

Using this system, biologically active TGFβ 1 in tissue culture supernatants,<br />

plasma, serum or urine can be quantitated in the range of 15.6–1,000pg/ml.<br />

Description: Anti-pS 473 Akt pAb is an affinity-purified polyclonal rabbit<br />

antibody. The antibody is purified using a phosphorylated peptide that<br />

corresponds to the phospho-S 473 form of Akt-1 and is useful for both Western<br />

blotting and immunocytochemistry.<br />

Features:<br />

• Specificity: The antibody is selective for the Ser 473 phosphorylated<br />

isoforms of Akt and does not show cross-reactivity with nonphosphorylated<br />

Akt.<br />

• Immunogen: Peptide from the singly phosphorylated Ser 473 region from<br />

the C-terminus of Akt-1 protein.<br />

Immunocytochemical staining of Akt in embryonic rat brain cells.<br />

Embryonic (day 17) rat brain cells were collected and treated with 20ng/ml<br />

each of EGF and FGF. Anti-pS 473 Akt pAb was used at a 1:50 dilution. Positive<br />

cells were visualized using a donkey anti-rabbit, Cy ® 3-conjugated secondary<br />

antibody. Nuclei were stained using DAPI. Protocols developed and performed<br />

at <strong>Promega</strong>.<br />
