Description: The PowerPlex ® CS7 System, Custom, is a multiplex STR assay<br />

for relationship testing and human identification. The PowerPlex ® CS7 System,<br />

Custom, allows co-amplification and three-color detection of seven STR loci,<br />

including LPL, F13B, FESFPS, F13A01, Penta D, Penta C and Penta E. All seven<br />

loci are amplified simultaneously in a single tube and analyzed in a single<br />

injection. The PowerPlex ® CS7 System, Custom, contains two loci, Penta D<br />

and Penta E, that overlap with the loci included in the PowerPlex ® 16 and<br />

PowerPlex ® 16 HS Systems. This feature allows the PowerPlex ® CS7 System,<br />

Custom, to be used as a confirmatory kit in paternity applications using the<br />

five unshared STR loci to supplement the genotype and increase the available<br />

information. The PowerPlex ® CS7 System, Custom, is compatible with the<br />

ABI PRISM ® 3100 and 3100-Avant Genetic Analyzers and Applied Biosystems<br />

3130 and 3130xl Genetic Analyzers. The PowerPlex ® CS7 System, Custom,<br />

provides all materials necessary to amplify STR regions of purified genomic DNA.<br />

Description: The PowerPlex ® S5 System is a mini-STR kit that allows<br />

co-amplification and detection of four STR markers (D18S51, D8S1179, TH01<br />

and FGA) plus Amelogenin. One primer specific for each of the Amelogenin,<br />

D18S51 and D8S1179 loci is labeled with fluorescein (FL), and one primer<br />

specific for each of the TH01 and FGA loci is labeled with 6-carboxy-4´,5´dichloro-2´,7´-dimethoxy-fluorescein<br />

(JOE). All five loci are amplified simultaneously<br />

in a single tube and analyzed in a single injection. The four STR loci are<br />

included in the CODIS and European databases. The amplicons for all loci are<br />

smaller than 260bp. The PowerPlex ® S5 System was the first <strong>Promega</strong> STR kit<br />

to include hot-start Taq DNA polymerase, which is included in the PowerPlex ®<br />

S5 5X Master Mix. The PowerPlex ® S5 System is primarily a screening tool but<br />

also can be used as a mini-STR casework kit.<br />

Features:<br />

• Sensitive: Generate full DNA profiles with as little as 50pg of DNA.<br />

• Easy to Use: The PowerPlex ® S5 System comes complete with premixed<br />

primer pairs, a master mix with Taq DNA polymerase and internal lane<br />

standard. The simplified thermal cycling protocol requires no ramping, and<br />

the system is compatible with a number of instrument platforms, including<br />

ABI PRISM ® 310, 3100 and 3100-Avant and Applied Biosystems 3130<br />

and 3130xl Genetic Analyzers.<br />

• Robust: The PowerPlex ® S5 System is more tolerant of DNA degradation<br />

and less sensitive to inhibitors. Full DNA profiles can be achieved in the<br />

presence of 130μM hematin, 200ng tannic acid or 150ng humic acid.<br />

• Automatic Assignment of Genotypes Available: Panels and bins text<br />

files are required for use with GeneMapper ® ID software and are available<br />

for download. The PowerTyper S5 Macro (available separately) facilitates<br />

data analysis, allowing automatic assignment of genotypes using the Genotyper<br />

® software. The PowerTyper Macros can be downloaded or ordered<br />

on CD-ROM.<br />
