Description: These Flexi ® Vectors are designed specifically for high-level<br />

expression of proteins in mammalian cells from the CMV promoter with or<br />

without a selectable marker. The pFN9A Vector provides Renilla luciferase,<br />

which may be used as a transfection control. The pFN9A Vector was designed<br />

to complement pGL4 firefly luciferase vectors when exogenous proteins (e.g.,<br />

a receptor of transcription factor) must be expressed for reporter assays.<br />

All inserts may be confirmed by cell-free expression with the TnT ® T7 Quick<br />

System (Cat.# L1170).<br />

Note: Flexi ® Vectors contain the lethal barnase gene to reduce background<br />

colonies without inserts during the subcloning procedure. Using the Flexi ® Vector<br />

Cloning System replaces the barnase gene with your insert. These<br />

vectors, as purchased, cannot be cultured in normal laboratory strains of<br />

E. coli without an insert.<br />

Description: The pALTER ® -MAX Vector is a 5,534bp plasmid. It contains the<br />

human cytomegalovirus (CMV) immediate-early enhancer/promoter region for<br />

strong, constitutive expression of cloned DNA inserts in a variety of mammalian<br />

cell types. The pALTER ® -MAX Vector as supplied is chloramphenicol-resistant<br />

and ampicillin-sensitive.<br />

Features:<br />

• High Expression: Protein expression driven by the human cytomegalovirus<br />

(CMV) immediate-early enhancer/promoter region allows high<br />

constitutive expression in a variety of cell lines. A chimeric intron composed<br />

of the 5´-splice site from the β-globin intron and the 3´-splice site from an<br />

IgG intron allows increased expression levels.<br />

• Efficient: SV40 late polyadenylation signal for efficient RNA processing. T7<br />

and T3 RNA polymerase promoters flank the multiple cloning site and allow<br />

RNA in vitro synthesis from sense or antisense strands.<br />

• Flexible: Restriction sites flanking the CMV enhancer (BglII and SgfI) and<br />

the CMV promoter (SgfI and I-PpoI) allow for easy replacement of these<br />

regions by alternative regulatory regions.<br />

Description: The pGEM ® -3Z Vector is intended for use as a standard cloning<br />

vector, as well as for the highly efficient synthesis of RNA in vitro. The vector<br />

carries the lacZ α-peptide and the multiple cloning region arrangement from<br />

pUC18 allowing selection of recombinants by blue/white screening. In addition,<br />

the vector contains both the SP6 and T7 RNA polymerase promoters flanking<br />

the multiple cloning region.<br />

The pGEM ® -3Z and pGEM ® -4Z Vectors are essentially identical except for the<br />

orientation of the SP6 and T7 promoters.<br />

Features:<br />

• Blue/White Screening: Allows the easy identification of recombinant<br />

clones.<br />

• Versatile: This vector can be used for standard cloning and in vitro<br />
