ABSTRACT<br />
Stability studies on 146S particles of inactivated and concentrated FMD virus<br />
antigens<br />
The aim of this study is to evaluate the effect of non-specific proteins which were<br />
concentrated together with virus inactivated by BEI and concentrated by cross-flow<br />
method on 146S amount during storage at 4 ºC and -20 ºC.<br />
At 12th month of study, 10 % antigen decrease was detected in concentrated type O<br />
vaccines stored at 4 o C, especially in group 3 and 4 (Group 3: Inactivated antigen 50% +<br />
Dulbecco PBS 48.5%+ Chloroform 3% + NaHCO3 1% Group 4: Inactivated antigen 50%<br />
+ Dulbecco PBS 48.5%+ Chloroform 0.5% + NaHCO3 1%) and 30 % of antigen decrease<br />
was detected in group 6 which was control group.<br />
All groups of concentrated type A vaccine antigens stored at 4 o C, after 3th month,<br />
15 and 90 % of antigen decreases were detected after month 3 and at 12th month<br />
respectiveley. 95 % of antigen decrease was detected in group 6 which was control group<br />
of type A antigens. However group 3 and 4 showed slightly higher stability than others<br />
which showed 60-70 % of antigen decrease at month 6.<br />
For all study groups of both type of antigens that stored -20 ºC, a sudden decline<br />
has been detected at month 3. For group 3, 4 and 6, relatively more stability have been<br />
detected.<br />
Thirty percent decrease for those stored at 4 C and 60 % for those stored at -20 C<br />
were observed in total protein levels of all groups of both antigen types. Overall, antigens<br />
stored at 4 ºC were more stable than those stored at -20 ºC. Of course -70 ºC and -196 ºC<br />
would be better for antigens storage. Although we did not try them in this study because of<br />
the volume problems that caused by large scale production. In this case, antigen<br />
preparations described in group 3 and 4 will be more useful for routine use.<br />
Today concentrated antigens are purified using PEG to get rid of irrelevant<br />
proteins. It was suggested that concentrated antigens may be stored at 4 o C up to two year<br />
by using further purification techniques.<br />
