3.ABSTRACT<br />
American foulbrood (AFB) caused by Paenibacillus larvae is a worldwideobserved<br />
bacterial disease in honey bee larvae. The present study aims to disease P. larvae,<br />
the disease cause, from honey and beeswax samples from suspected hives by direct PCR<br />
and culture. AF6 and AF7, the most sensitive pair of primers, were used to identify the<br />
DNA of P. larvae.<br />
A total of 100 suspected honey and beeswax samples collected from 25 different<br />
enterprises were examined. In 8 samples, P. larvae were identified through culture method.<br />
For isolation of P. larvae, Brain-Heart Infusion (BHI) agar enriched with 0.2 g/l thiamine<br />
hydrochloride, blood agar and blood agar supplemented with 3µg/ml nalidixic acid were<br />
