OBiE-Cr: Fxogenous progester<strong>on</strong>e has been shown to reduce brain edema and<br />
ischeemia-Induced cell damage and to improve physiological and neurological<br />
functi<strong>on</strong> during the early stage of focal cerebral Ischemla. In the present study,<br />
the authors assessed the neuroprotective potential of progester<strong>on</strong>e during the<br />
late stage of ischemia in a transient middle cerebral artery (MCA) occlusi<strong>on</strong><br />
model In the rat. METHODS: Forty-eight male sp<strong>on</strong>taneously hypertensive rats<br />
were randomly assigned to six groups. Progester<strong>on</strong>e was dissolved in dimethyl<br />
sulfoxide (DMSO). In four groups of rats, the dissolved progester<strong>on</strong>e (4 mg/kg<br />
or 8 mg/kg) was administered for 2 or 7 days after Ischemia. In two c<strong>on</strong>trol<br />
groups DMSO was administered for 2 or 7 days after ischemia. Occlusi<strong>on</strong> of the<br />
MCA was Induced by Inserti<strong>on</strong> Of an Intraluminal suture, and reperfusl<strong>on</strong> was<br />
accomplished by withdrawal of the suture. Treatment was initiated <strong>on</strong><br />
reperfusi<strong>on</strong>, which followed 2 hours of MCA occlusi<strong>on</strong>, and c<strong>on</strong>tinued <strong>on</strong>ce a day.<br />
Lesi<strong>on</strong> volume, neurological deficit; and body weight loss were measured 2 or 7<br />
days after ischemla, depending <strong>on</strong> fhe animal group. Treatment with a high dose .<br />
of progester<strong>on</strong>e (8 mg/kg) resulted In reducti<strong>on</strong>s In lesi<strong>on</strong> size, neurological<br />
deficits, and body weight, compared with c<strong>on</strong>trol rats. CONCLUSIONS:<br />
Administrati<strong>on</strong> of progester<strong>on</strong>e to male rats 2 hours after MCA occlusi<strong>on</strong> reduces<br />
Ischemic brain damage and improves neurological deficit even 7 days after<br />
Ischemia.<br />
