INTENDED USE & SUMMARY<br />
Urine based tests for multiple drugs of abuse range from simple immunoassay tests<br />
to complex analytical procedures. The speed and sensitivity of immunoassays have<br />
made them the most widely accepted method to test urine for multiple drugs of<br />
abuse. The "DIAQUICK" <strong>DOA</strong> <strong>Multi</strong>-6 <strong>Cup</strong> is a lateral flow chromatographic<br />
immunoassay for the qualitative detection of following drugs of abuse without the<br />
need of additional instruments:<br />

This test will also detect other related compounds, please refer to the Analytical<br />
Specificity table in this package insert. This assay provides only a preliminary<br />
analytical test result. A more specific alternate chemical method must be used in<br />
order to obtain a confirmed analytical result. Gas chromatography/mass<br />
spectrometry (GC/MS) is the preferred confirmatory method. Clinical consideration<br />
and professional judgment should be applied to any drug of abuse test result,<br />
particularly when preliminary positive results are used. For in vitro diagnostic use<br />
only.<br />
TEST PRINCIPLE<br />
The "DIAQUICK" <strong>DOA</strong> <strong>Multi</strong>-6 <strong>Cup</strong> is an immunoassay based on the principle of<br />
competitive binding. Drugs which may be present in the urine specimen compete<br />
against their respective drug conjugate for binding sites on their specific antibody.<br />
During testing, a urine specimen migrates upward by capillary action. A drug, if<br />
present in the urine specimen below its cut-off concentration, will not saturate the<br />
binding sites of its specific antibody coated on the particles. The antibody coated<br />
particles will then be captured by the immobilized drug conjugate and a visible<br />
colored line will show up in the test line region of the specific drug strip. The colored<br />
line will not form in the test line region if the drug level is above its cut-off<br />
concentration because it will saturate all the binding sites of the antibody coated on<br />
the particles. A drug-positive urine specimen will not generate a colored line in the<br />
specific test line region of the strip because of drug competition, while a drugnegative<br />
urine specimen or a specimen containing a drug concentration less than<br />
the cut-off will generate a line in the test line region. To serve as a procedural<br />
control, a colored line will always appear at the control line region indicating that<br />
proper volume of specimen has been added and membrane wicking has occurred.<br />
REAGENTS<br />
Each test line in the test panel contains mouse monoclonal antibody-coupled<br />
particles and corresponding drug-protein conjugates. A goat antibody is employed<br />
in each control line.<br />
PRECAUTIONS<br />
• For medical and other professional in vitro diagnostic use only.<br />
• Do not use after the expiration date.<br />
• The test device should remain in the sealed pouch until use.<br />
• All specimens should be considered as potentially hazardous and handled in the<br />
same manner as an infectious agent.<br />
• The used test panel should be discarded according to local regulations.<br />
STORAGE AND STABILITY<br />
Store as packaged in the sealed pouch either at room temperature or refrigerated<br />
(2-30°C). The test panel is stable through the expiration date printed on the sealed<br />
pouch. The test panel must remain in the sealed pouch until use. DO NOT<br />
FREEZE. Do not use beyond the expiration date.<br />
SPECIMEN COLLECTION AND PREPARATION<br />
Urine collected at any time of the day may be used. Urine specimens exhibiting<br />
visible precipitates should be centrifuged, filtered, or allowed to settle to obtain a<br />
clear supernatant for testing.<br />
SPECIMEN STORAGE<br />
Urine specimens may be stored at 2-8°C for up to 48 hours prior to assay. For<br />
prolonged storage, specimens may be frozen and stored below -20°C. Frozen<br />
specimens should be thawed and mixed before testing.<br />

DIRECTIONS FOR USE<br />
Allow the test panel, urine specimen, and/or controls to equilibrate to room<br />
temperature (15-30°C) prior to testing.<br />
1. Bring the pouch to room temperature before opening it. Remove the cup from<br />
the sealed pouch and use it as soon as possible.<br />
2. Remove the key by twisting it from the center of the cup cap.<br />
3. Collect specimen in the cup and secure the cap tightly by pressing down on the<br />
pull tab until an audible click is heard.<br />
4. Check the temperature label (Temp Label) up to 4 minutes after specimen<br />
collection. A green color will appear to indicate the temperature of the urine<br />
specimen. The proper range for an unadulterated specimen is 32-38°C (90-<br />
100°F).<br />
5. Date and initial the security seal label, then place it over the cap.<br />
6. Place the cup on a flat surface and push the key into the socket of the cup to<br />
initiate the test. Start the timer.<br />
7. Remove the peel off label covering the test results and wait for the colored<br />
line(s) to appear. Read results at 5 minutes. Do not interpret results after 10<br />
minutes.<br />
INTERPRETATION OF RESULTS<br />
NEGATIVE:* A colored line in the control line region (C) and a colored line in the<br />
test line region (T) for a specific drug indicate a negative result. This indicates that<br />
the drug concentration in the urine specimen is below the designated cut-off level<br />
for that specific drug.<br />
*NOTE: The shade of color in the test line region (T) may vary, but it should be<br />
considered negative whenever there is even a faint colored line.<br />
POSITIVE: A colored line in the control line region (C) but no line in the test line<br />
region (T) for a specific drug indicates a positive result. This indicates that the drug<br />
concentration in the urine specimen exceeds the designated cut-off for that specific<br />
drug.<br />
INVALID: Control line fails to appear. Insufficient specimen volume or incorrect<br />
procedural techniques are the most likely reasons for control line failure. Review the<br />
procedure and repeat the test using a new test cup. If the problem persists,<br />
discontinue using the lot immediately and contact your local distributor.<br />
QUALITY CONTROL<br />
A procedural control is included in the test. A colored line appearing in the control<br />
line region (C) is considered an internal procedural control. It confirms sufficient<br />
specimen volume, adequate membrane wicking and correct procedural technique.<br />
Control standards are not supplied with this kit; however, it is recommended that<br />
positive and negative controls be tested as good laboratory practice to confirm the<br />
test procedure and to verify proper test performance.<br />
LIMITATIONS<br />
The "DIAQUICK" <strong>DOA</strong> <strong>Multi</strong>-6 <strong>Cup</strong> provides only a preliminary analytical result. A<br />
more specific chemical method must be used to obtain a confirmed result. Gas<br />
chromatography/mass spectrometry (GC/MS) is the preferred confirmatory method.<br />
2. It is possible that technical or procedural errors, as well as other interfering<br />
substances in the urine specimen may cause erroneous results.<br />
3. Adulterants, such as bleach and/or alum, in urine specimens may produce<br />
erroneous results regardless of the analytical method used. If adulteration is<br />
suspected, the test should be repeated with another urine specimen.<br />
4. A positive result indicates presence of the drug or its metabolites but does not<br />
indicate level of intoxication, administration route or concentration in urine.<br />
5. A negative result may not necessarily indicate drug-free urine. Negative results<br />
can be obtained when drug is present but below the cut-off level of the test.<br />
6. The test does not distinguish between drugs of abuse and certain medications.<br />
7. A positive result might be obtained from certain foods or food supplements.<br />

Accuracy<br />
A side-by-side comparison was conducted using the "DIAQUICK" <strong>DOA</strong> <strong>Multi</strong>-6 <strong>Cup</strong><br />
and commercially available drug rapid tests. Testing was performed on<br />
approximately 300 specimens. Presumptive positive results were confirmed by<br />
GC/MS. The following results were tabulated:<br />

A drug-free urine pool was spiked with drugs to the concentrations at ± 50% cut-off<br />

Cross-Reactivity<br />
A study was conducted to determine the cross-reactivity of the test with compounds<br />
in either drug-free urine or Amphetamine, Benzodiazepines, Cocaine, Marihuana,<br />
Methamphetamine, Morphine positive urine. The following compounds show no<br />
cross-reactivity when tested with the "DIAQUICK" <strong>DOA</strong> <strong>Multi</strong>-6 <strong>Cup</strong> (Urine) at a<br />
concentration of 100 μg/mL.<br />

ANWENDUNG & KLINISCHE BEDEUTUNG<br />
Drogentests für Urin für den gleichzeitigen Nachweis von verschiedenen Drogen<br />
gibt es in unterschiedlichsten Ausführungen und reichen von einfacheren<br />
Immunoassays bis hin zu komplexen analytischen Methoden. Die schnell<br />
erzielbaren Ergebnisse und die hohe Sensitivität haben sie zu den meist<br />
verbreitetsten Tests für den gleichzeitigen Nachweis von mehreren Drogen in Urin<br />
gemacht. Der "DIAQUICK" <strong>DOA</strong> <strong>Multi</strong>-6 <strong>Cup</strong> ist ein immunchromatographischer<br />
Membranschnelltest für den qualitativen Nachweis der folgenden Drogen ohne<br />
zusätzlich benötigte Instrumente:<br />

Mit diesem Test werden auch andere, verwandte Substanzen miterfasst. Die<br />
notwendigen Informationen können aus der Tabelle “analytische Spezifität“<br />
entnommen werden. Dieser Test liefert nur ein vorläufiges analytisches Ergebnis.<br />
Eine andere, spezifischere chemische Methode muss für ein endgültiges Ergebnis<br />
herangezogen werden. Gaschromatographie/Massenspektrometrie (GC/MS) ist die<br />
Bestätigungsmethode der Wahl. Die Berücksichtigung klinischer Aspekte und eine<br />
professionelle Beurteilung der Ergebnisse sollten für jeden Drogentest angewandt<br />
werden, besonders dann, wenn nur vorläufige positive Testergebnisse vorliegen.<br />

TESTPRINZIP<br />
Der "DIAQUICK" <strong>DOA</strong> <strong>Multi</strong>-6 <strong>Cup</strong> ist ein Immunoassay, der auf dem Prinzip der<br />
kompetitiven Bindung basiert. Drogen, die im Urin enthalten sein könnten,<br />
verdrängen das Drogen-Goldkonjugat von den Bindungsstellen am Antikörper.<br />
Während der Testdurchführung wandert die Urinprobe durch die Kapillarkräfte die<br />
Membran entlang aufwärts. Sind Drogen unterhalb der entsprechenden cut-off<br />
Konzentrationen in der Urinprobe enthalten, so werden die Bindungsstellen der<br />
spezifischen antikörperbeschichteten Partikel nicht abgesättigt. Die<br />
antikörperbeschichteten Partikel werden dann vom immobilisierten Drogenkonjugat<br />
gebunden und es bildet sich im Testfenster des entsprechenden Teststreifens eine<br />
farbige Testlinie aus. Die gefärbte Testlinie erscheint nicht, wenn die<br />
Drogenkonzentration über der entsprechenden cut-off Konzentration liegt, weil alle<br />
Bindungsstellen der mit Antikörpern beschichteten Partikel mit Drogenmolekülen<br />
besetzt sind. Daher führt eine drogenpositive Urinprobe aufgrund der<br />
Verdrängungsreaktion nicht zu einer gefärbten Testlinie auf dem jeweiligen<br />
Teststreifen, während eine negative Urinprobe bzw. eine Urinprobe, die Drogen<br />
unterhalb der cut-off Konzentration enthält, zu einer sichtbaren Testlinie führt. Die<br />
Kontrolllinie muss sich unabhängig von der Anwesenheit des Analyts immer bilden.<br />
Sie dient als interne Kontrolle und zeigt an, dass ausreichend Probe aufgetragen<br />
wurde und dass der Test funktioniert.<br />
REAGENZIEN<br />
Jede Testlinie auf der Testkarte enthält Partikel, die mit monoklonalen<br />
Mausantikörpern sind und das korrespondierende Drogen-Protein-Konjugat. Ein<br />
Ziegenantikörper ist in den Kontrolllinien enthalten.<br />
VORSICHTSMAßNAHMEN<br />
• Nur für medizinische und andere professionelle in-vitro-diagnostische Zwecke<br />
geeignet! Nicht nach Überschreiten des Ablaufdatums verwenden!<br />
• Der Test sollte erst unmittelbar vor der Testdurchführung aus der Verpackung<br />
entnommen werden!<br />
• Alle Proben sollten als potenziell infektiös betrachtet werden und so behandelt<br />
werden, wie infektiöses Material.<br />
• Die verwendeten Tests müssen entsprechend den regional gültigen<br />
Bestimmungen entsorgt werden.<br />
LAGERUNG & HALTBARKEIT<br />
Der Test sollte bei 2-30°C oder im Kühlschrank gelagert werden. Der Test ist bis<br />
zum auf der Verpackung angegebenen Ablaufdatum haltbar. Der Test muss in der<br />
versiegelten Verpackung bis kurz vor der Testdurchführung gelagert werden. Nicht<br />
einfrieren! Ablaufdatum beachten!<br />
PROBENGEWINNUNG UND PROBENVORBEREITUNG<br />
Der für diesen Test verwendete Urin kann zu jeder Tageszeit gesammelt werden.<br />
Urinproben mit sichtbarem Präzipitat sollten zentrifugiert oder gefiltert werden.<br />
Geeignet ist auch das Absetzenlassen von Trübungen.<br />
PROBENLAGERUNG<br />
Urinproben können bei 2-8°C bis zu 48 Stunden aufbewahrt werden. Für eine<br />
längere Lagerung ist eine Temperatur von -20°C erforderlich. Eingefrorene Proben<br />
müssen vor der Testdurchführung komplett aufgetaut werden.<br />

TESTDURCHFÜHRUNG<br />
Vor Durchführung des Tests sind die Patientenproben sowie das Kontroll- und<br />
Referenzmaterial auf Raumtemperatur (20°-30°C) zu bringen.<br />
1. Der Becher wird aus der versiegelten Verpackung entnommen und so bald wie<br />
möglich verwendet.<br />
2. Entfernen Sie den Schlüssel, indem Sie ihn mit einer drehenden Bewegung<br />
aus der Mitte des Deckels herausziehen.<br />
3. Die Probe wird im Becher gesammelt und der Deckel wird dicht verschlossen,<br />
indem die Lasche nach unten gedrückt wird, bis ein hörbares Klicken<br />
vernommen wird.<br />
4. Das Temperaturlabel sollte innerhalb von 4 Minuten nach der<br />
Probengewinnung kontrolliert werden.<br />
5. Es erscheint eine grüne Farbe und zeigt damit die Temperatur der Urinprobe<br />
an. Der Normalbereich bei nicht verfälschtem Urin liegt zwischen 32 und 38°C.<br />
6. Beschriften Sie die Sicherheitsetikette und kleben Sie sie über die<br />
Verschlusskappe.<br />
7. Stellen Sie den Becher auf eine flache Unterlage und drücken Sie den<br />
Schlüssel in die Buchse, damit der Test gestartet wird. Nun wird die Stopuhr<br />
gestartet.<br />
8. Um die Ergebnisse ablesen zu können, muss zuvor die Abziehetikette, die<br />
zunächst das Ergebnisfenster verdeckt, abgezogen werden. Die Ergebnisse<br />
sollten nach 5 Minuten abgelesen werden. Die Ergebnisse nicht später als<br />
nach 10 Minuten ablesen.<br />
INTERPRETATION DER ERGEBNISSE<br />
NEGATIV:* Es erscheinen zwei farbige Linien für einen Testparameter. Eine in der<br />
Kontrollregion (C) und eine in der Testregion (T). Ein negatives Resultat zeigt an, dass<br />
keine Droge in der Probe ist, bzw. deren Konzentration unter der Nachweisgrenze des<br />
Tests liegt.<br />
*Achtung: Eine sehr schwache Linie in der Testregion (T) zeigt an, dass die<br />
Drogenkonzentration der Probe, nahe dem Cut-off Wert des Testsystems liegt.<br />
Auf jeden Fall ist der Test beim Erscheinen einer, wenn auch noch so schwachen Linie,<br />
als negativ zu werten und gegebenenfalls mit einer spezifischeren Methode nochmals<br />
zu untersuchen.<br />
POSITIV: Eine farbige Linie erscheint in der Kontrollzone (C). Es bildet sich keine<br />
Linie in der Testregion (T) auf dem entsprechenden Teststreifen. Ein positives<br />
Resultat zeigt an, dass die Drogenkonzentration in der Probe über dem<br />
entsprechenden Cut-off Wert des Testes liegt.<br />
UNGÜLTIG: Weder in der Kontrollzone (C) noch in der Testzone (T) eines<br />
Testparameters erscheint eine gefärbte Linie. Dies zeigt eine falsche Testdurchführung,<br />
ein Versagen der Testkarte oder eine Verfälschung der Probe an. Auf jeden Fall den<br />
Test mit einer neuen Testkarte wiederholen. Bei neuerlichem Versagen des Tests bitte<br />
notieren Sie die Lot. Nummer und das Ablaufdatum des Tests und kontaktieren Sie den<br />
Händler.<br />
QUALITÄTSKONTROLLE<br />
Eine interne Kontrolle zur Überprüfung der richtigen Durchführung ist auf der<br />
Testeinrichtung beinhaltet. Eine rote Linie in der Kontrollregion (C) dient zur<br />
Kontrolle der richtigen Durchführung. Die rote Linie bestätigt, dass genügend<br />
Probenvolumen aufgebracht und der Test richtig durchgeführt wurde.<br />
Kontrollstandards sind im Testkit nicht enthalten. Trotzdem wird empfohlen,<br />
negative und positive Proben zur Überprüfung des Tests heranzuziehen um in<br />
Übereinstimmung mit den Good Laboratory Practice Richtlinien die Funktion des<br />
Tests zu verifizieren zu können.<br />
EINSCHRÄNKUNGEN DER METHODE<br />
Der "DIAQUICK" <strong>DOA</strong> <strong>Multi</strong>-6 <strong>Cup</strong> liefert nur ein vorläufiges analytisches Ergebnis.<br />
Eine andere, spezifischere chemische Methode muss für ein endgültiges Ergebnis<br />
herangezogen werden. Gaschromatographie/Massenspektrometrie (GC/NS) ist die<br />
Bestätigungsmethode der Wahl.<br />
1. Es besteht die Möglichkeit, dass technische Fehler auftreten oder Fehler bei der<br />
Testdurchführung gemacht werden. Außerdem können im Urin vorhandene<br />
störende Substanzen zu fehlerhaften Ergebnissen führen.<br />
2. Verfälschende Substanzen in Urin, wie Bleichmittel oder Alaun, können<br />
unabhängig von der verwendeten Methode zu fehlerhaften Ergebnissen führen.<br />
Wenn eine Verfälschung vermutet wird, so sollte der Test mit einer neuen Probe<br />
wiederholt werden.<br />

3. Ein positives Ergebnis zeigt an, dass Drogenspuren im Urin nachgewiesen<br />
werden konnten. Dieses Ergebnis sagt aber nichts bezüglich einer möglichen<br />
Intoxikation, der Art der Verabreichung oder der Konzentration im Urin aus.<br />
4. Ein negatives Ergebnis bedeutet nicht zwingend, dass im Urin keine<br />
Drogenspuren vorhanden sind. Negative Ergebnisse können auch dann zu<br />
beobachten sein, wenn die Konzentration unterhalb des jeweiligen Cut-off Wertes<br />
liegt.<br />
5. Der Test kann nicht zwischen bestimmten Medikamenten und Drogen<br />
unterscheiden.<br />
6. Ein positives Ergebnis kann durch bestimmte Supplemente oder Nahrungsmittel<br />
ausgelöst werden.<br />
TESTCHARAKTERISTIK<br />
Genauigkeit<br />
Es wurde ein Vergleich zwischen dem "DIAQUICK" <strong>DOA</strong> <strong>Multi</strong>-6 <strong>Cup</strong> und einem<br />
kommerziell erhältlichen Schnelltest durchgeführt. Es wurden ca. 300 Proben für<br />
diese Evaluierung verwendet. Vorläufige positive Ergebnisse wurden mit GC/MS<br />
bestätigt. Die Ergebnisse sind der folgenden Tabelle zu entnehmen.<br />

Ein drogenfreier Urin wurde mit Drogen versetzt (Konzentrationen: ± 50% cut-off<br />

Kreuzreaktivität<br />
Es wurde eine Studie durchgeführt, um die Kreuzreaktivität des Testes mit<br />
verschiedenen Substanzen in drogenfreiem Urin, und Amphetamin-,<br />
Benzodiazepin-, Cocain-, Marihuana-, Methamphetamin-, Morphin-positivem Urin<br />
zu bestimmen. Die folgenden Verbindungen zeigten keine Kreuzreaktivität mit dem<br />
"DIAQUICK" <strong>DOA</strong> <strong>Multi</strong>-6 <strong>Cup</strong> bis zu einer Konzentration von 100 μg/mL:<br />
